In order to understand the similarity or difference of inorganic As species uptake and transport related to phosphorus in As-hyperaccumulator, uptake and transport of arsenate (As(Ⅴ)) and arsenite (As(Ⅲ)) were studied using Pteris vittata L. under sand culture. Higher concentrations of phosphate were found to inhibit accumulation of arsenate and arsenite in the fronds of P. vittata. The reduction in As accumulation was greater in old fronds than in young fronds, and relatively weak in root and rhizome. Moderate increases, from 0.05 to 0.3 mmol/L, in phosphate reduced uptake of As(Ⅲ) more than As(Ⅴ), while the reverse was observed at high concentrations of phosphate (≥ 1.0 mmol/L). Phosphate apparently reduced As transport and the proportion of As accumulated in fronds of P. vittata when As was supplied as As(Ⅴ). It may in part be due to competition between phosphorus and As(Ⅴ) during transport. In contrast, phosphate had a much smaller effect on As transport when the As was supplied as As(Ⅲ). Therefore, the results from present experiments indicates that a higher concentration of phosphate suppressed As accumulation and transport in P. vittata, especially in the fronds, when exposure to As(Ⅴ); but the suppression of phosphate to As transport in the root or rhizome may be insignificant when P. vittata when exposure to As(Ⅲ) under sand culture conditions. The finding will help to understand the interaction of P and As during their uptake process in P. vittata.  相似文献

为探究锌(Zn)对水稻镉(Cd)累积的影响及其根表铁膜所发挥的作用,选取Cd高累积型水稻品种中9优547(简称"Z547")和Cd低累积型水稻品种金优402(简称"J402"),采用温室水培试验,研究0、2、5、10、15和20 μmol/L等6个Zn浓度下水稻幼苗对Cd的累积效应,以及不同浓度Zn处理对根表铁膜生成量的影响.结果表明:①随着c(Zn)的增加,Z547和J402水稻幼苗生物量均呈先增后减的趋势,分别在c(Zn)为2和10 μmol/L时达到最大值.②Z547和J402水稻幼苗中w(Cd)均呈先降后增的趋势,分别在c(Zn)为5和2 μmol/L时达到最小值;当水稻幼苗中w(Cd)达到最小值时,Z547根和地上部中w(Cd)分别为31.65和11.47 mg/kg,J402根和地上部中w(Cd)分别为22.58和14.36 mg/kg.③不同浓度Zn处理下水稻幼苗各部位中w(Cd)均与根表铁膜中w(Mn)、w(Fe)、w(Fe+Mn)呈显著正相关,高铁膜处理水稻幼苗中w(Cd)显著高于低铁膜处理,表明根表铁膜生成量的增加会促进Cd在水稻幼苗中的累积.研究显示,当c(Zn)较低时,c(Zn)的增加会抑制水稻幼苗对Cd的累积;当c(Zn)较高时,c(Zn)的增加会促进水稻幼苗对Cd的累积,而Zn可通过控制根表铁膜的生成来影响水稻幼苗对Cd的累积.   相似文献

来源于土壤和灌溉水的砷在水稻根表及其体内的富集特性   总被引:6,自引:2,他引:4  
采用土壤-玻璃珠联合培养的方式,选择2个氧化能力不同的水稻品种YY-1、94D-64(品系)和采自浙江富阳的土壤(砷的本底值为13.8 mg·kg-1),并设灌溉清水和含砷水2个处理(即在分蘖期、拔节期、孕穗期、开花期和灌浆期5个生育阶段灌溉含砷污水,随灌溉水进入土壤中砷的浓度为3.2mg·kg-1),研究了砷在土壤-根表铁氧化物-水稻系统中的累积规律以及土壤和灌溉水对水稻秸秆和籽粒富集砷的贡献程度.结果表明,水稻的秸秆生物量及其籽粒产量并没有受到不同来源砷的显著影响;灌溉含砷水处理的两品系水稻根表铁氧化物沉积的数量(YY-1∶196 g·kg-1,94D-64∶75.8 g·kg-1)高于对照(YY-1∶175g·kg-1,94D-64∶60.1 g·kg-1),但差异不显著.然而,在水稻5个不同的生育期灌溉含砷水均显著增加了砷在其根表及其体内不同部位的富集(94D-64籽粒中砷含量除外).没有灌溉含砷水的对照其秸秆和籽粒中累积的砷来源于土壤,而砷处理的水稻其秸秆和籽粒中富集的砷则来源于土壤和灌溉的含砷水.土壤对YY-1和94D-64秸秆中富集砷的贡献率分别为76.5%和71.O%,灌溉水的贡献率分别为23.5%和29.0%,2个水稻品系之间没有明显差异.YY-1籽粒中的砷66.4%来源于土壤,33.6%来源于含砷灌溉水,灌溉水对该品系籽粒中砷的富集贡献率较高.另一品系94D-64籽粒中砷84.8%来源于土壤,15.2%由灌溉水贡献,灌溉水对此品系籽粒累积砷的贡献率较低.来源于土壤和灌溉水的砷在水稻籽粒中的富集没有超出我国的国家食品卫生标准(0.7 mg·kg-1).  相似文献

A hydroponic experiment was conducted to investigate the effects of arsenic (As) stress on growth, nutrition and As uptake, and speciation in shoots and roots of winter wheat (Triticum aestivum L.). Winter wheat has high tolerance to As. Most As is accumulated in the roots, and an As concentration of 4,421 mg/kg was observed at a solution concentration of 20 mg/L As. Arsenic concentrations in roots were approximately 40-100 times greater than those in shoots. Arsenic in winter wheat roots and shoots occurred as both As^3+ and As^5+ species, although As^3+ was the main species in winter wheat tissues. Arsenic significantly decreased the biomass of winter wheat shoots and roots and affected absorption and transport of micro- and macro-elements in winter wheat tissue. Arsenic treatment significantly increased the concentrations of total Magnesium (Mg) and calcium (Ca) in shoots and enhanced the transport of Mg and Ca from roots to shoots but decreased potassium (K), nitrogen (N), and phosphorus (P) concentrations in both shoots and roots, particularly the concentration of P. Concentrations of iron, copper, and zinc in winter wheat shoots were negatively related to As rates, with correlation coefficients (R^2) of 0.93, 0.94, and 0.97, respectively.  相似文献

宁波南沙港网箱养殖水域营养状况评价及生物修复策略   总被引:1,自引:0,他引:1  
根据2007年1月-2007年11月4个航次对宁波南沙港网箱养殖水域水体营养盐(NO2--N、NO3--N、NH 4+-N和PO34--P)含量的监测结果,利用营养指数E法对水体的富营养化状态进行了评价。根据网箱养殖系统物质平衡方程和现场调查实验数据,估算了南沙港网箱养殖水域的氮磷污染负荷,以溶解态氮作为平衡指标,估算了鱼藻类合理的配比模式。结果表明,南沙港水体中NO2--N、NO3--N、NH4+-N、PO4-P含量年变化范围分别为0.26μmol/L~4.86μmol/L,33.93μmol/L~82.86μmol/L,0.59μmol/L~7.86μmol/L,1.47μmol/L~2.97μmol/L,不同养殖区之间营养盐含量差异不显著(p〈0.05);营养指数E值年变化范围介于2.41~15.99之间,表明该港水体处于重度富营养化状态;养殖区N/P值的年平均值为32.95,表明南沙港海区是一个氮过剩的养殖系统。南沙港网箱养殖水域氮、磷的负荷量分别为70.43 t/a和5.96 t/a,其中溶解态氮、磷负荷量分别为49.3 t/a和2.38 t/a;南沙港的藻类养殖面积需要在现有基础上再扩大0.44倍,达到107.52 ha,才可以与网箱养殖系统的氮磷污染负荷构成平衡;网箱养殖个数与藻类养殖面积(ha)的合理配比约为1∶0.027。  相似文献

In the present study, arsenate(As(V)) and phosphate(P(V)) interactions were investigated in growth, uptake and RNA content in yeast(Saccharomyces cerevisiae). Yeast grew slowly with As(V) concentrations increasing in the medium. However, the maximal population density was almost the same among different As(V) treatments. It was in the late log phase that yeast growth was aug- mented by low As(V), which was maybe due to the fact that methionine metabolism was stressed by vitamin B6 deprivation, so As(V) treatments did not affect maximal population density. However, with P (V) concentrations increasing, the maximal population density increased. Therefore, the maximal population density was determined by P (V) concentrations in the medium but not by As (V) concentrations in the medium. Ycflp(a tonoplast transpor) transports As(GS)3 into the vacuole, but arsenic(As) remaining in the thalli was 1.27% with As(V) exposure for 60 h, from which it can be speculated that the percentage of As transported into vacuole should be lower than 1.27%. However, the percentage of As pumped out of cell was 71.49% with As (V) exposure for 68 h. Although two pathways (extrusion and sequestration) were involved in As detoxification in yeast, the extrusion pathway played a major role in As detoxification. RNA content was the highest in the early-log phase and was reduced by As(V).  相似文献

Arsenic (As) speciation for the phytoremediation by the Chinese brake fern was studied. In particular, the mechanism of how plants induce compounds containing thiol (SH) and proteins by As exposure in terms of the relationship between As and phosphate uptaken into plant cells was examined. Pteris vittata callus could efficiently reduce As(V) to As(III) by the rapid introduction of reductase and synthesize thiols leading to phytochelatins production. Furthermore, Pteris vittata could control phosphate concentration in the cells corresponding to the concentration of arsenite and arsenate. To our best knowledge, this is the first report to show the mechanisms of such high As tolerance of Pteris vittata using their callus in terms of in vitro approach for the analysis of As speciation and metabolism route.  相似文献

铜、砷单一及复合污染对黑藻的毒性效应   总被引:2,自引:0,他引:2  
通过室内模拟试验,研究了铜(Cu)、五价砷〔As(Ⅴ)〕单一及复合污染对黑藻〔Hydrilla verticillata (L.f.) Royle〕生长、富集Cu和As的能力以及对超氧化物歧化酶(SOD)活性的影响.毒性测试结果表明,Cu和As(Ⅴ)对黑藻的4 d-EC50分别为2和12 μmol/L,单一Cu对黑藻的毒性远高于As(Ⅴ). 复合污染试验结果表明,对于5 μmol/L As(Ⅴ)处理组,低浓度Cu〔c(Cu)<2 μmol/L〕的加入,促进了植物生长,降低了植物体内的As总量〔w(As)〕,同时降低了植物体中占主要部分的三价砷的含量(w〔As(Ⅲ)〕). 对于Cu处理组,As(Ⅴ)的加入同样抑制了黑藻对Cu的富集,SOD活性也高于单一Cu处理,可见在低浓度Cu处理下,Cu和As(Ⅴ)之间存在拮抗作用;而高浓度Cu〔c(Cu)≥2 μmol/L〕的加入,虽然植物对Cu和As(Ⅴ)的富集与单一Cu和As(Ⅴ)处理相比均显著降低,但植物生长受到抑制,SOD活性与单一处理相比也显著降低(P<0.05),因此高浓度Cu处理下,二者对黑藻毒性起协同作用. 总之,黑藻具有应用于处理低浓度Cu和As(Ⅴ)单一及复合污染水体的潜力.   相似文献

The growth, cellular total lipids, bioaccumulation amount, and bioaccumulation factors (BAFs) of 2,4,4 ′ -tribromodiphenyl ether (BDE28), 2,2 ′ ,4,4 ′ -tetrabromodiphenyl ether (BDE47), and 2,2 ′ ,4,4 ′ ,5-pentabromodiphenyl ether (BDE99) in a semi-continuous culture of Prorocentrum donghaiense were studied in relation to nitrate (0, 128, and 512 μmol/L) and phosphate (0, 8, and 32 μmol/L) concentrations. The BDE28, BDE47, and BDE99 content per cell under 0 μmol N/L were 3.77 × 10 6 , 3.95 × 10 6 , and 4.32 × 10 6 ng/cell, respectively, which were significantly higher than those under 128 and 512 μmol N/L. A nearly 5-fold increase in polybrominated diphenyl ether (PBDE) content per algal cell was found between 0 and 8 μmol P/L and between 8 and 32 μmol P/L. With increasing N and P concentrations, the PBDE content per volume of algal culture and the accumulation percentage of available PBDEs declined slightly. The BAFs for the PBDEs based on lipids showed that the logBAF lip under 0 μmol N/L was higher than those under 128 and 512 μmol N/L. The logBAF lip under 0 μmol P/L was higher than that under 8 μmol P/L but lower than that under 32 μmol P/L. Correlation analysis indicated a significant negative correlation between nutrient concentration and cellular total lipids, as well as the PBDE content per cell. The results indicate that different N and P concentrations change the total lipids content of P. donghaiense, thereby resulting in varying PBDE accumulation.  相似文献

Arsenic (As) is a pervasive and ubiquitous environmental toxin that has created catastrophic human health problems world-wide. Chlamydomonas reinhardtii is a unicellular green alga, which exists ubiquitously in freshwater aquatic systems. Arsenic metabolism processes of this alga through arsenate reduction and sequent store and efflux were investigated. When supplied with 10 mol/L arsenate, arsenic speciation analysis showed that arsenite concentration increased from 5.7 to 15.7 mg/kg dry weight during a 7-day period, accounting for 18%–24% of the total As in alga. When treated with different levels of arsenate (10, 20, 30, 40, 50 mol/L) for 7 days, the arsenite concentration increased with increasing external arsenate concentrations, the proportion of arsenite was up to 23%–28% of the total As in alga. In efflux experiments, both arsenate and arsenite could be found in the efflux solutions. Additionally, the efflux of arsenate was more than that of arsenite. Furthermore, two arsenate reductase genes of C. reinhardtii (CrACR2s) were cloned and expressed in Escherichia coli strain WC3110 (ΔarsC) for the first time. The abilities of both CrACR2s genes to complement the arsenatesensitive strain were examined. CrACR2.1 restored arsenate resistance at 0.8 mmol/L. However, CrACR2.2 showed much less ability to complement. The gene products were demonstrated to reduce arsenate to arsenite in vivo. In agreement with the complementation results, CrACR2.1 showed higher reduction ability than CrACR2.2, when treated with 0.4 mmol/L arsenate for 16 hr incubation.  相似文献

This study investigates arsenic (As) accumulation and tolerance of duckweed Spirodela polyrhiza L: and its potential for As phytofiltration. S. polyrhiza was able to survive in high concentration of As(V) solution: The EC50 values ( SE) based on the external As(V) were (181.66 20.12) mol/L. It accumulated (999 95) mg As/kg dw when exposed in 320 mol/L As(V) solution for one week, and was able to take up appropriately 400 mg As/kg dw in tissues without a significant biomass loss. The EC50 values (the e ective concentration of As(V) in the nutrient solution that caused a 50% inhibition on biomass production) was (866 68) mg/kg dw for the tissues, indicating that S. polyrhiza had a high capability of As accumulation and tolerance. The uptake kinetic parameters Vmax was (55.33 2.24) nmol/(g dw min) and Km was (0.144 0.011) mmol/L. Within 72 hr, S. polyrhiza decreased As concentration in the solution from 190 to 113 ng/mL with a removal rate of 41%. The study suggested that this floating aquatic plant has some potential for As phytofiltration in contaminated water bodies or paddy soils.  相似文献

砷污染是全球环境热点问题之一,土壤砷污染治理是一个急需解决的难题.随着不同砷超富集植物的发现,植物修复技术因其投资和维护成本低、不易造成二次污染等优点而成为国际学术界研究的热点和前沿领域.深入理解超富集植物体内砷代谢和富集机制是有效利用植物修复技术来治理砷污染土壤的关键.近年来,植物超富集砷机制的研究取得了很大进展.已有的研究结果证明,蜈蚣草(P.vittata)对砷的吸收和转运能力显著高于非超富集植物,转运到地上部的砷主要储存在羽叶细胞的液胞中;蜈蚣草具有很强的抗氧化胁迫能力和五价砷(As(Ⅴ))还原能力;菌根共生有利于蜈蚣草的生长和砷的富集.有关植物超富集砷机制的分子生物学研究也取得了可喜的进展.砷酸盐还原酶基因(PvACR2)和植络素合成酶基因(PvPCS1)都被克隆并表征,cTPI同源基因(PV4-8)也被证明具有将As(Ⅴ)还原成三价砷(As(Ⅲ))的功能.尽管植物超富集砷机制的研究取得了迅速进展,但至今学术界仍没有全面理解为什么这些蕨类植物具有超富集砷的功能,相关功能基因、酶和转运蛋白的研究有待进一步深入.  相似文献

砷(As)污染对作物生长有毒害作用,导致作物品质和产量降低,因此亟需采取安全有效的策略以减轻其毒害.采用水培法,通过荧光探针定位技术、差速离心技术和傅里叶红外光谱技术(FTIR)研究外源茉莉酸(JA)对水稻幼苗As积累及抗逆应答的影响.结果表明,外源添加JA使水稻地下部和地上部的As含量分别降低31.4%和51.4%,显著降低细胞壁和细胞液中的As含量.JA改变亚细胞组分中As的分布比例,使细胞壁中As的分布比例提高16.4%,细胞液中As的分布比例降低17.3%,增强细胞壁对As的固定,减少细胞液中的As含量.此外,JA提高根细胞中SOD、CAT、GSH和PEPC的水平,降低H2 O2和MDA含量,减轻脂质过氧化损伤,调节碳氮代谢,缓解As毒害.研究为水稻As污染防治提供了新思路.  相似文献

Contamination of irrigation water represents a major constraint to Bangladesh agriculture, resulting in elevated levels in the terrestrial systems. Lux bacterial biosensor technology has previously been used to measure the toxicity of metals in various environmental matrices. While arbuscular mycorrhizal fungi have their most significant effect on phosphorus uptake, but showed alleviated metal toxicity to the host plant. The study examined the effects of arsenic and inoculation with an arbuscular mycorrhizal fungus, Glomus mosseae, on lentil (Lens culinaris L. cv. Titore). Plants were grown with and without arbuscular mycorrhizal inoculum for 9 weeks in a sand and terra-green mixture (50:50, V/V) and watered with five levels of arsenic (0, 1, 2, 5, 10 mg As/L arsenate). The results showed that arsenic addition above 1 mg/L significantly reduced percentage of mycorrhizal root infection. On further analysis a close relationship was established with the vegetative and reproductive properties of lentil (L. culinaris) plants compared to the percentage bioluminescence of the soil leachate. However, arbuscular mycorrhizal fungal inoculation reduced arsenic concentration in roots and shoots. Higher concentrations of arsenic (5, 10 mg As/L arsenate) reduced the mycorrhizal efficiency to increase phosphorus content and nitrogen fixation. Therefore, this study showed that increased concentration of arsenic in irrigation water had direct implications to the lentil (L. culinaris) plants overall performance. Moreover the use of bioassay demonstrated that mycorrhiza and clay particle reduced arsenic bioavailability in soil.  相似文献

以系统评价有机磷农药在水域生态系统风险为目标,采用批次培养的方法研究了草甘膦和毒死蜱两种有机磷农药在高浓度下的毒性效应、低浓度下的刺激效应及微量条件下的无机磷替代效应.结果表明,草甘膦和毒死蜱对四尾栅藻(Scenedesmus quadricanda)的96h EC50值的分别为12.98,2.02μmol/L.低浓度草甘膦(0.014~1.35μmol/L)条件下细胞密度和最大光能转化效率表现出了显著的刺激作用,最大促进效应分别为13%和6.0%;草甘膦(0.40~0.67μmol/L)表现出显著的无机磷替代效应,而毒死蜱处理组未观察到刺激作用和替代效应.有机磷农药对微藻种群变化的作用方式以低浓度下的刺激作用为主,同时可以引发无机磷不足时某些可利用有机磷种类微藻的种群增长.  相似文献

Pot and field experiments were conducted to investigate the effects of water regimes on the speciation and accumulation of arsenic (As) and cadmium (Cd) in Brazilian upland rice growing in soils polluted with both As and Cd. In the pot experiment constant and intermittent flooding treatments gave 3-16 times higher As concentrations in soil solution than did aerobic conditions but Cd showed the opposite trend. Compared to arsenate, there were more marked changes in the arsenite concentrations in the soil solution as water management shifted, and therefore arsenite concentrations dominated the As speciation and bioavailability in the soil. In the field experiment As concentrations in the rice grains increased from0.14 to 0.21 mg/kg while Cd concentrations decreased from 0.21 to 0.02 mg/kg with increasing irrigation ranging from aerobic to constantly flooding conditions. Among the various water regimes the conventional irrigation treatment produced the highest rice grain yield of 6.29 tons/ha. The As speciation analysis reveals that the accumulation of dimethylarsinic acid (from 11.3% to 61.7%) made a greater contribution to the increase in total As in brown rice in the intermittent and constant flooding treatments compared to the intermittent-aerobic treatment. Thus, water management exerted opposite effects on Cd and As speciation and bioavailability in the soil and consequently on their accumulation in the upland rice. Special care is required when irrigation regime methods are employed to mitigate the accumulation of metal(loid)s in the grain of rice grown in soils polluted with both As and Cd.  相似文献

A pot experiment was conducted to investigate the effects of root oxidation on arsenic (As) dynamics in the rhizosphere and As sequestration on rice roots. There were significant differences (P 〈 0.05) in pH values between rhizosphere and non-rhizosphere soils, with pH 5.68-6.16 in the rhizosphere and 6.30-6.37 in non-rhizosphere soils as well as differences in redox potentials (P 〈 0.05). Percentage arsenite was lower (4%-16%) in rhizosphere soil solutions from rice genotypes with higher radial oxygen loss (ROL) compared with genotypes with lower ROL (P 〈 0.05). Arsenic concentrations in iron plaque and rice straw were significantly negatively correlated (R = -0.60, P 〈 0.05). Genotypes with higher ROL (TD71 and Yinjingmanzhau) had significantly (P 〈 0.001) lower total As in rice grains (1.35 and 0.96 mg/kg, respectively) compared with genotypes with lower ROL (IAPAR9, 1.68 mg/kg; Nanyangzhan 2.24 mg/kg) in the As treatment, as well as lower inorganic As (P 〈 0.05). The present study showed that genotypes with higher ROL could oxidize more arsenite in rhizosphere soils, and induce more Fe plaque formation, which subsequently sequestered more As. This reduced As uptake in aboveground plant tissues and also reduced inorganic As accumulation in rice grains. The study has contributed to further understanding the mechanisms whereby ROL influences As uptake and accumulation in rice.  相似文献
