Toxin content (fmol cell–1) and a suite of elemental and macromolecular variables were measured in batch cultures of the dinoflagellatesAlexandrium fundyense, A. tamarense andAlexandrium sp. from the southern New England region, USA. A different perspective was provided by semicontinuous cultures which revealed sustained, steady-state physiological adaptations by cells to N and P limitation. Two types of variability were investigated. In batch culture, changes in nutrient availability with time caused growth stage variability in toxin content, which often peaked in mid-exponential growth. A second type of variability that could be superimposed on growth stage differences is best exemplified by the high toxin content of cells grown at suboptimal temperatures. Calculations of the net rate of toxin production (R tox ; fmol cell–1 d–1) for these different culture treatments and modes made it possible to separate the dynamics of toxin production from cell division. Over a wide range of growth rates, cells produced toxin at rates approximating those needed to replace losses to daughter cells during division. The exception to this direct proportionality was with P limitation, which was associated with a dramatic increase in the rate of toxin production as cells stopped dividing due to nutrient limitation in batch culture. Growth stage variability in batch culture thus reflects small imbalances (generally within a factor of two) between the specific rates of toxin production and cell division. N limitation and CO2 depletion both affect pathways involved in toxin synthesis before those needed for cell division; P limitation does the opposite. The patterns of toxin accumulation were the same as for major cellular metabolites or elemental pools. The highest rates of toxin production appear to result from an increased availability of arginine (Arg) within the cell, due to either a lack of competition for this amino acid from pathways involved in cell division or to increased de novo synthesis. There were no significant changes in toxin content with either acclimated growth at elevated salinity, or with short term increases or decreases of salinity. These results demonstrate that toxin production is a complex process which, under some conditions, is closely coupled to growth rate; under other conditions, these processes are completely uncoupled. Explanations for the observed variability probably relate to pool sizes of important metabolites and to the differential response of key biochemical reactions to these pool sizes and to environmental conditions.  相似文献

Activities of lactate dehydrogenase (LDH), pyruvate kinase (PK), malate dehydrogenase (MDH) and citrate synthase (CS) were measured in the white skeletal muscle of marine fishes having different depths of occurrence and different feeding and locomotory strategies. There were significant depth-related differences in the two glycolytic enzymes, LDH and PK. LDH activity was most variable, and differed by 3 orders of magnitude between the most active shallow-living species and certain deep-sea fishes likely to have only minimal capacities for active locomotion. Superimposed on the depth-related patterns was a high degree of interspecific variation (up to 20-fold) in enzymic activity among species from any given range of depth of occurrence. Variation of both LDH and PK activities, noted for shallow- and deep-living fishes, seems to be largely accounted for by differences in feeding habits and locomotory performance. Active pelagic swimmers have much higher activities of LDH and PK than, for example, deep-living sit-and-wait predators. Benthopelagic fishes like rattails and the sablefish have the highest activities found among deep-living fishes, suggesting that these species engage in relatively active food-searching behavior compared to most other deep-sea fishes. The activity of CS, an enzyme of the citric acid cycle and an indicator of aerobic metabolism, varied little among species. Thus, the large interspecific variation in glycolytic potential (LDH and PK) among species is not associated with a similar variation in aerobic metabolism of white muscle. The much higher and more variable activity of MDH relative to CS suggests that, in addition to its function in the citric acid cycle, MDH may play an important role in redox balance in fish white muscle. In a comparison of white muscle composition between the shallow- and deep-living species, water content did not differ significantly, but protein content was significantly higher in shallow- than in deep-living fishes (211 and 130 mg g-1 wet wt of muscle, respectively). The differences in muscle protein content are small relative to the differences between shallow- and deep-living species in LDH, PK and MDH activities. Thus, depthrelated differences in muscle enzymic activity are caused by factors other than enzyme dilution. Enzyme activities (LDH, PK and CS) in brain tissue were relatively constant among species regardless of depth of occurrence or feeding and locomotory habits. Habitat and lifestyle do not seem to influence the demands for overall metabolic function in brain. The utility of muscle enzymic activity data for making predictions about the ecological characteristics of difficult-to-observe, deep-living, fishes is discussed.  相似文献

On the basis of theoretical considerations, population-based nutrition surveys of 30 clusters of 30 children should provide reasonably valid estimates of the prevalence of malnutrition with at least 95 per cent confidence that the estimated prevalence differs from the true value by no more than 5 per cent. In areas of famine in Africa, where an urgent need often exists for rapid nutritional assessment to determine the extent and severity of the problem, visiting 30 sites is often logistically difficult. To determine the effects of using fewer than 30 clusters on the validity and precision of the estimated level of undernutrition, we used data from the 1983 Swaziland National Nutrition Survey and from rapid nutrition surveys performed in 1984 and 1985 in Burkina Faso, Guinea, and Niger. Fewer than 30 clusters may result in point prevalence estimates that differ dramatically from the true prevalence and, in most instances, are less precise. In contrast, little is gained by collecting more than 30 clusters. In summary, around 30 clusters provides relatively valid and precise estimates of the prevalence of undernutrition, and every effort should be made to obtain the logistic support required to study this number of clusters.  相似文献

ABSTRACT

Natural history films use technological mediations to frame aspects of nature so as to communicate information, in part through engendering particular viewer affects. As an entertainment industry embedded in capitalist social relations and concerned with competition for finance and ratings, natural history film-making is also a search for “the money shot” – associated with extremes including rarity, sensational behavior, and otherwise un(fore)seen views. I highlight this sensationalizing impetus through ethnographic fieldwork conducted at the biannual UK Wildscreen film festival in 2010. Here, wildlife films were frequently presented as action dramas with a rhythm of anticipation, climax and satisfaction. I argue that, through stimulating significantly disconnective affects, such framing may work against composition of a caring ecocultural ethics that entwines human with more-than-human natures and futures; and that this tendency parallels the similarly disconnecting effects documented for pornographic film. In contrast, I engage with the differently constructive frames guiding the low budget, open access, activist film Green, which, perhaps paradoxically given the thrust of much of the natural history film industry, won the prestigious WWF Gold Panda Award at Wildscreen 2010. I follow framing theorist George Lakoff to emphasize that since cognition is both embodied and embedded in diverse inter-relationships, affective registers generating mimetic connection are as significant in communicating information regarding “the environment” as the text and words by which nature might be framed. I conclude that attention to affective registers and embodied (dis)connections in natural history film may enhance a turning of capitalist spectacle against itself, so as to work more effectively in service to the composition of abundant socionatural futures.  相似文献

Preplant soil fumigation is an important pest management practice in coastal California strawberry production regions. Potential atmospheric emissions of fumigants from field treatment, however, have drawn intensive environmental and human health concerns; increasingly stringent regulations on fumigant use have spurred research on low-emission application techniques. The objectives of this research were to determine the effects of a new low-permeability film, commonly known as totally impermeable film (TIF), on fumigant emissions and on fumigant distribution in soil. A 50/50 mixture of 1,3-dichloropropene (1,3-D) and chloropicrin (CP) was shank-applied at 314 kg ha in two location-separate field plots (0.4 ha each) in Ventura County, California, in fall 2009. One plot was surface-covered with standard polyethylene (PE) film, and the other was covered with TIF immediately after fumigant application. Data collection included emissions, soil-gas phase concentration profile, air concentration under the film, and soil residuals of the applied fumigants. Peak emission flux of 1,3-D and CP from the TIF field was substantially lower than from the PE field. Total through-film emission loss was 2% for 1,3-D and <1% for CP from the TIF field during a 6-d film covering period, compared with 43% for 1,3-D and 12% for CP from the PE field. However, on film-cutting, greater retention of 1,3-D in the TIF field resulted in a much higher emission surge compared with the PE field, while CP emissions were fairly low in both fields. Higher concentrations and a more uniform distribution in the soil profile for 1,3-D and CP were observed under the TIF compared with the PE film, suggesting that the TIF may allow growers to achieve satisfactory pest control with lower fumigant rates. The surging 1,3-D emissions after film-cutting could result in high exposure risks to workers and bystanders and must be addressed with additional mitigation measures.  相似文献

Introduction

Previous studies have shown that increased risk in darkness is particularly great for pedestrian crashes, suggesting that attempts to improve headlighting should focus on factors that likely influence those crashes. The current analysis was designed to provide information about how details of pedestrian crashes may differ between daylight and darkness. Method: All pedestrian crashes that occurred in daylight or dark conditions in Michigan during 2004 were analyzed in terms of the variables included in the State of Michigan crash database. Additional analysis of the narratives and diagrams in police accident reports was performed for a subset of 400 of those crashes—200 sampled from daylight and 200 sampled from darkness. Results: Several differences were found that appear to be related to the characteristic asymmetry of low-beam headlamps, which (in the United States) distributes more light on the passenger's side than the driver's side of the vehicle. These results provide preliminary quantification of the how the photometric differences between the right and left sides of typical headlamps may affect pedestrian crash risk.

Impact on Industry

The results suggest that efforts to provide supplemental forward vehicle lighting in turns may have safety benefits for pedestrians.  相似文献

In this study we demonstrate the sensitivity of swimming behavior and predator-escape responses of nauplii of the estuarine copepod Eurytemora affinis to sublethal doses of Cu and Cd. Behavior was generally altered at metal doses below those affecting growth rates or survival of the copepods. Swimming velocities of Cu-dosed nauplii were different from controls at all concentrations of Cu tested (10–50 g l-1 total Cu) after 24- to 48-h exposure, whereas development rate of nauplii was significantly reduced only after 96 h at 25 g l-1. The 96 h LC50 for Cu was approximately 30 g l-1 Cu. Naupliar swimming velocity was also affected by Cd. Swimming speeds were reduced after 24 h at 130 g l-1, and development was slowed after 48 h at 116 g Cd l-1. The 96-h LC50 was >120 g l-1. Little is known of the adaptive role of specific motile behaviors in the success of larval copepods. We investigated the relationship of swimming speed to predator — prey interactions of the nauplii using both real and simulated predators. Nauplii exposed to Cu for 24 h were observed to be generally hyperactive, a condition which could increase their encounter frequency with predators. Reduced numbers of escape responses of nauplii to a simulated predator, another indication of increased vulnerability to predation, were observed only after 48-h exposure to Cu. Nevertheless, feeding rates of non-dosed larval striped bass on dosed nauplii (24 h at 25 g Cu l-1) were significantly higher than on control nauplii. We study the potentially unnecessary costs imposed by strict performance standards for forest restoration of surface coal mines in the Appalachian region under the Surface Mining Control and Reclamation Act of 1977 (SMCRA) that can vary widely across states. Both the unnecessary private costs to the mine operator and costs to society (social costs) are reported for two performance standards, a ground cover requirement, and a seedling survival target. These standards are examined using numerical analyses under a range of site productivity class and market conditions. We show that a strict (90%) ground cover standard may produce an unnecessary private cost of more than 700/ha and a social cost ranging from700/ha and a social cost ranging from 428/ha to 710/ha, as compared with a 70 standard, may produce an unnecessary private cost of approximately710/ha, as compared with a 70% standard. A strict tree survival standard of 1235 trees/ha, as compared with the more typical 1087 trees/ha standard, may produce an unnecessary private cost of approximately 200/ha, and a social cost in the range of 120 to120 to 208/ha. We conclude that strict performance standards may impose substantial unnecessary private costs and social costs, that strict performance standards may be discouraging the choice of forestry as a post-mining land use, and that opportunities exist for reform of reforestation performance standards. Our study provides a basis for evaluating tradeoffs between regulatory efficiency and optimal reforestation effort.  相似文献
