Five extracellular PHB depolymerases of bacteria isolated from various sources were purified to electrophoretic homogeneity and compared with known extracellular PHB depolymerase fromAlcaligenes faecalis T1. The molecular mass of these enzymes were all around 40–50 kDa. Nonionic detergent, diisopropylfluorophosphate and dithiothreitol inhibited the PHB depolymerase activity of all these enzymes. Trypsin abolished PHB depolymerase activity, but not theD-3-hydroxybutyric acid dimer hydrolase activity of all the enzymes. These results showed that the basic properties of these PHB depolymerases resemble those of theA. faecalis T1 enzyme. Analysis ofN-terminal amino acid sequence of the purified enzymes revealed that these enzymes includingA. faecalis T1 enzyme fall into three groups.  相似文献

A poly(3-hydroxybutyrate) (PHB) depolymerase was purified from a fungus, Penicillium funiculosum (IFO6345), with phenyl-Toyopearl and its properties were compared with those of other PHB depolymerases. The molecular mass of the purified enzyme was estimated at about 33 kDa by sodium dodecyl sulfate (SDS) polyacrylamide gel electrophoresis. The pH optimum and pI were 6.5 and 6.5, respectively. The purified protein showed affinity to Con A-Sepharose, indicating that it is a glycoprotein. Diisopropylfluorophosphate and dithiothreitol inhibited the depolymerase activity completely. The N-terminal amino acid sequence of the purified enzyme was TALPAFNVNPNSVSVSGLSSGGYMAAQL, which contained a lipase box sequence. This purified enzyme is one of the extracellular PHB depolymerase which belong to serine esterase. The purified enzyme showed relatively strong hydrolytic activity against 3-hydroxybutyrate oligomers compared with its PHB-degrading activity. PHB-binding experiments showed that P. funiculosum depolymerase has the weakest affinity for PHB of all the depolymerases examined.  相似文献

A soil–plant–air continuum multilayer model was used to numerically simulate canopy net assimilation (An), evapotranspiration (ET), and soil moisture in a deciduous teak plantation in a dry tropical climate of northern Thailand to examine the influence of soil drought on An. The timings of leaf flush and the end of the canopy duration period (CDP) were also investigated from the perspective of the temporal positive carbon gain. Two numerical experiments with different seasonal patterns of leaf area index (LAI) were carried out using above-canopy hydrometeorological data as input data. The first experiment involved seasonally varying LAI estimated based on time-series of radiative transmittance through the canopy, and the second experiment applied an annually constant LAI. The first simulation captured the measured seasonal changes in soil surface moisture; the simulated transpiration agreed with seasonal changes in heat pulse velocity, corresponding to the water use of individual trees, and the simulated An became slightly negative. However, in the second simulation, An became negative in the dry season because the decline in stomatal conductance due to severe soil drought limited the assimilation, and the simultaneous increase in leaf temperature increased dark respiration. Thus, these experiments revealed that the leaflessness in the dry season is reasonable for carbon gain and emphasized the unfavorable soil water status for carbon gain in the dry season. Examining the duration of positive An (DPA) in the second simulation showed that the start of the longest DPA (LDPA) in a year approached the timing of leaf flush in the teak plantation after the spring equinox. On the other hand, the end appeared earlier than that of all CDPs. This result is consistent with the sap flow stopping earlier than the complete leaf fall, implying that the carbon assimilation period ends before the completion of defoliation. The model sensitivity analysis in the second simulation suggests that a smaller LAI and slower maximum rate of carboxylation likely extend the LDPA because soil water from the surface to rooting depth is maintained longer at levels adequate for carbon gain by decreased canopy transpiration. The experiments also suggest that lower soil hydraulic conductivity and deeper rooting depth can postpone the end of the LDPA by increasing soil water retention and the soil water capacity, respectively.  相似文献

In order to understand the residue levels of organochlorine compounds (OCs) and their accumulation patterns in wildlife inhabiting Chubu region, Japan, the concentrations of polychlorinated biphenyls (PCBs), hexachlorocyclohexane isomers (HCHs), DDT compounds (DDTs) and hexachlorobenzene (HCB) were measured in 8 species of terrestrial mammals and 10 species of birds. In view of feeding habits, the contamination levels of OCs were found to be higher in omnivorous mammals than in herbivorous ones, and in fish-eating ones and raptores than in omnivorous birds. In fox and dog, PCB-180 (2, 2′, 3, 4, 4′, 5, 5′-heptachlorobiphenyl) was the most dominant PCB congener, while in the other species PCB-153 (2, 2′, 4, 4′, 5, 5′-hexachlorobiphenyl) was the most persistent. The ratios of lower chlorinated PCB congeners (tri- to tetra-) to total PCBs were larger in fish-eating birds than in the other birds. The results indicate that the compositions of PCB congeners would reflect the differences of feeding habits and xenobiotic metabolizing systems among each species.  相似文献
