Samples of the scleractinian coral Pocillopora damicornis were collected from six sites located around four islands in the Ryukyu Archipelago, southern Japan, and subjected to allozyme electrophoresis. Seven polymorphic loci were examined for their allelic patterns. The ratio of observed to expected genotypic diversity (0.30 < G o :G e  < 0.64), the ratio of the observed number of genotypes to the number of individuals (0.47 < N g :N i  < 0.75), and deviations from Hardy–Weinberg equilibrium indicated that asexual reproduction plays a major role in the maintenance of established populations. However, populations were not completely dominated by a single or a few clones, and most clones were represented by only a few individual samples. The high frequency of typhoons in the region suggests that, in P. damicornis, fragmentation caused through occasional exposure to powerful waves is a major mode of asexual reproduction, but asexual production of planulae may also be contributing to the maintenance of populations. A significant genetic differentiation (F ST) was found between the six populations examined (0.027 < F ST < 0.092, average F ST = 0.056). The moderate gene flow is discussed according to characteristics of the larval stage of the species, and to circulation patterns in the region. This study aimed to assess Japan’s recent “local production for local consumption” (LPLC) movement, with a special focus on vegetables in the Osaka city region of central Japan. After collecting statistics and spatial data, we conducted a multi-scale analysis of vegetable production and consumption along with the associated energy consumption, using geographical information system software at three spatial scales along the vegetable flow paths: national, regional, and local. Vegetables consumed in the Osaka city region came from prefectures throughout Japan, and we mapped the foodshed within this region at 1-km spatial resolution, as well as the distribution of farmland, farmers’ and other markets. We also conducted a scenario analysis for reduced energy consumption through organic farming and the utilization of abandoned farmland near a city to replace food imports from distant areas. We found that the large majority of vegetables consumed in the Osaka city region currently come from remote prefectures, and that this is associated with a high level of energy consumption. Inside the Osaka city region, peri-urban vegetable farming contributes to regional vegetable provision, resulting in an approximately 70 % production/consumption ratio within an 80-km radius of the urban center; if all of the area of abandoned farmland were restored to production, this ratio would increase to approximately 75 %. Organic farming activities that bring together farmers and urbanites are emerging in many parts of the study area, contributing to increased LPLC. Scenario analysis suggested that a decrease of more than 1 × 106 GJ of energy inputs could be achieved through wider adoption of local organic farming for local consumption and complete utilization of abandoned farmland in the Osaka city region.  相似文献

The aim of this study was to examine the effects of ultraviolet A (UV-A) irradiation-induced damage on cultured macrophage RAW 264.7 cells and determine which components produced these manifestations. RAW 264.7 cells were irradiated with 365 nm UV-A using a light-emitting diode (LED). Cell viability and damage were determined using a calcein-AM and propidium iodide dual-staining assay and lactate dehydrogenase leakage, respectively. Intracellular reactive oxygen species (ROS) were measured by H2DCF-DA. The components of ROS in each medium were measured using an electron paramagnetic resonance (EPR) spectrometer in the presence of 5,5-dimethyl-1-pyrroline-N-oxide (DMPO) and 2,2,5,5-tetramethyl-3-pyrroline-3-carboxamide (TPC). While UV-A irradiation for 2 min significantly suppressed cell growth, LDH leakage did not occur. Addition of N-acetyl cysteine restored inhibition of cell proliferation, and reduced intracellular ROS levels. The EPR signal in the presence of TPC increased with time but was decreased by sodium azide. In addition, a typical EPR spectrum was obtained in the presence of DMPO, indicating the presence of a hydroxyradical. The spectrum was diminished by L-histidine. Data suggest that ROS generated in cells or culture medium by UV-A irradiation is predominantly singlet oxygen, and this singlet oxygen suppressed cell proliferation.  相似文献

The major source of dioxin impurities in Japan in the past was agrochemical formulations; more recently, it has been exhaust from waste incinerators. To examine the environmental and genetic factors that influence blood dioxin concentration, we investigated the relationship among dioxin concentrations, dietary habits and cytochorome P450 1A1 (CYP1A1) polymorphisms (MspI type and Ile-Val type) in Japanese fishermen and farmers, including also a group of office workers as controls. The mean dioxin concentrations in the fishermen, the farmers and the controls were 161369, 79079 and 100500 pg/g fat, respectively. The elevated dioxin concentration with polychlorinated dibenzo-p-dioxins, polychlorinated dibenzofurans and coplanar-PCBs found in the fishermen may be due to the frequent consumption of fish; no such relationship was found both in the farmers and the controls. We found that the concentrations of congeners found as impurities in certain chemicals such as those previously used in agriculture showed no significant differences among the three groups; we consider it unlikely that the farmers would be directly exposed to dioxins from such chemicals. Thus, it is probable that the primary route of dioxin exposure in the Japanese population is through the food chain via fish consumption, regardless of occupation. No meaningful relationship between blood dioxin concentration and CYP1A1 polymorphisms was found in this study, although there was a significant difference between the concentration of total non-ortho-PCBs in genotypes A and B. Further studies on more subjects, including those of genotype C, are needed to confirm the relationship between blood dioxin concentrations and MspI polymorphisms.  相似文献

Development of an echinothurid sea urchin is described for the first time; eggs of Asthenosoma ijimai (Yoshiwara) have been fertilized in the laboratory, and development has been followed light-microscopically up to the early juvenile stage. The eggs, which are orange and float, are 1.2 mm in diameter, by far the largest echinoid eggs on record. The embryos, larvae and early juveniles are lecithotrophic, and no exogenous food is needed for development. The embryonic stages include a remarkable biscuit-shaped late blastula, which has never been described for any echinoid before. The larva, with its para-arms unsupported by skeletal ossicles, is unlike the echinoid prism or pluteus and more resembles the early bipinnaria or early auricularia of asteroids and holothurians, respectively. All stages through late larvae float just beneath the surface until settlement occurs during the third and fourth weeks at 20°C. Metamorphosis from late larva to juvenile is gradual and no part of the larval body appears to be cast off.This paper is dedicated to Professor Juro Ishida on the occasion of his seventieth birthday.  相似文献

Male genitalia show rapid and divergent evolution. It is rarely determined whether variation in male genital morphology influences male reproductive success. Male damselflies possess a unique aedeagus with a re-curved head and spiny lateral processes, and most females have two sperm storage organs, a spherical bursa copulatrix and a tubular spermatheca. Previous studies have indicated that the re-curved head may remove bursal sperm, whereas the lateral processes remove spermathecal sperm. However, we need more direct evidence of these functions. We compared sperm number in female sperm storage organs by interrupting copulation to examine sperm removal by the male. In Calopteryx cornelia, males removed almost all bursal sperm but only partially removed spermathecal sperm. In contrast, females of Mnais pruinosa store sperm primarily in the bursa, and males removed only bursal sperm. To examine the functions of male spiny lateral processes, we compared mating behaviour between control and experimental males from which we removed (cut) the lateral processes. In C. cornelia, cutting of the lateral processes resulted in a decreased number of abdominal movements during copulation and no removal of spermathecal sperm. The amount of bursal sperm removed during copulation also decreased in experimental males compared to the unmanipulated males. However, in M. pruinosa, the experimental removal of male lateral processes did not decrease the abdominal movements and little affected the removal of bursal sperm. Inter-specific differences between C. cornelia and M. pruinosa may be caused by variation in the strategies of female sperm storage.  相似文献
