Abstract

Ambient air particulate matter less than 2.5 μm in aerodynamic diameter (PM2.5) samples were collected during summer and autumn using a Staplex high-volume air sampler. They were later extracted with dichloromethane in a Soxhlet apparatus. Polyaromatic hydrocarbon (PAH) content in extracts was determined by the high-performance liquid chromatography technique using fluorescence detection, whereas the nitro-PAH content was determined by gas chromatography using mass detection. Four Salmonella typhimurium strains (TA98, TA100, YG1041, and YG1042) were used in assays conducted with and without metabolic activation. The extracts were also tested with the SOS chromotest supplied by Environmental Biodetection Products Incorporated. The obtained results confirmed the Salmonella assay and the SOS chromotest usability for the purpose of atmospheric pollution monitoring within an urban agglomeration. The atmospheric pollution extracts under examination differed among each other regarding total content and percentage of individual compounds, depending on the season of sampling. The highest total PAH content and the highest nitro-PAH content in the tested samples as well as the most extensive range of detected compounds were found in the autumn season (heating season). The highest mutagenicity was noted for PM2.5 samples collected in autumn. The high values of mutagenicity ratios and induction factors were obtained from assays carried out with and without metabolic activation, which is an argument for the presence of promutagens and direct mutagens. The YG1041 strain proved to be the most effective in detection of mutagenicity of the suspended dust extracts because of its notably high sensitivity to nitro-aromatic compounds. The SOS chromotest was very sensitive to a large spectrum of genotoxic air pollutants and showed a high degree of similarity with the results of the Salmonella assay. In comparison with the frequently used Ames test, the SOS chromotest enables quick analysis of the genotoxic effects of samples using only one tester strain. In addition, its miniaturized design decreases the consumption of tested samples.  相似文献

Organisms in the environment are exposed to a mixture of pollutants. Therefore the purpose of this study was to analyze the mutagenicity of organic and inorganic responses in two fractions of particulates (TSP and PM2.5) and extracts (organic and aqueous). The mutagenicity of organic and aqueous particulate matter extracts from urban-industrial and urban-residential areas was evaluated by Salmonella/microsome assay, through the microsuspension method, using strain TA98 with and without liver metabolization. Additionally, strains YG1021 and YG1024 (nitro-sensitive) were used for organic extracts. Aqueous extracts presented negative responses for mutagenesis and cytotoxicity was detected in 50% of the samples. In these extracts the presence of potential bioavailable metals was identified. All organic extracts presented mutagens with a higher potential associated with PM2.5. This study presents a first characterization of PM2.5 in Brazil, through the Salmonella/microsome assay. The evaluation strategy detected the anthropic influence of groups of compounds characteristically found in urban and industrial areas, even in samples with PM values in accordance with quality standards. Thus, the use of a genotoxic approach in areas under different anthropic influences will favor the adoption of preventive measures in the health/environment relation.  相似文献

Bottom sediment and suspended sediment samples from Hamilton Harbour (western Lake Ontario) and from a major tributary were profiled using a bioassay-directed fractionation approach. Sample extracts were fractionated using an alumina/Sephadex gel clean-up procedure to afford non-polar aromatic fractions which were characterized using chemical analyses and the Ames/microsome bacterial assay in Salmonella typhimurium strains YG1025 with the addition of oxidative metabolism (S9), and YG1024 without S9. Non-polar aromatic fractions of selected samples were separated by normal phase HPLC into 1-min fractions which were subjected to bioassay analyses. The bioassays using strain YG1025+S9, a TA100-type strain, were performed to assess genotoxicity arising from the presence of polycyclic aromatic hydrocarbons (PAH). Fractions which exhibited mutagenic activity contained PAH with molecular masses of 252, 276 and 278 amu; these fractions contained over 80% of the genotoxicity attributable to PAH. Individual compounds identified using Gas Chromatography-Mass Spectrometry analyses in these active fractions included benzo[a]pyrene, indeno[cd]pyrene and dibenz[a,h]anthracene. The YG1025+S9 mutagenic activity profiles were similar for all samples. Mutagenic activity profiles generated using strain YG1024-S9, a TA98-type strain sensitive to compounds characteristic of mobile source emissions, were very different. The mutagenic activities in strain YG1024-S9 were greatest for harbour-suspended sediment samples collected from sites impacted by a major tributary. Suspended sediments collected near areas known to contain high levels of coal tar-contamination in the bottom sediments contained higher levels of genotoxic PAH than suspended sediments collected from other areas of the harbour.  相似文献

In the routine São Paulo state (Brazil) surface water quality-monitoring program, which includes the Salmonella microsome mutagenicity assay as one of its parameters, a river where water is taken and treated for drinking water purposes has repeatedly shown mutagenic activity. A textile dyeing facility employing azo-type dyes was the only identifiable source of mutagenic compounds. We extracted the river and drinking water samples with XAD4 at neutral and acidic pH and with blue rayon, which selectively adsorbs polycyclic compounds. We tested the industrial effluent, raw, and treated water and sediment samples with YG1041 and YG1042 and compared the results with the TA98 and TA100 strains. The elevated mutagenicity detected with YG-strains suggested that nitroaromatics and/or aromatic amines were causing the mutagenicity detected in the samples analyzed. Positive responses for the blue rayon extracts indicated that mutagenic polycyclic compounds were present in the water samples analyzed. The mutagen or mixture of mutagens present in the effluent and water samples cause mainly frameshift mutations and are positive with and without metabolic activation. The Salmonella assay combined with different extraction procedures proved to be very useful in the identification of the origin of the pollution and in the identification of the classes of chemical compounds causing the mutagenic activity in the river analyzed.  相似文献

Airborne particulate matter, PM(10) and PM(2.5), are associated with a range of health effects including lung cancer. Their complex organic fraction contains genotoxic and carcinogenic compounds such as polycyclic aromatic hydrocarbons (PAHs) and their derivatives. This study evaluates the genotoxicity of the PM(10) and PM(2.5) organic extracts that were sampled in the framework of a personal exposure study in three French metropolitan areas (Paris, Rouen and Strasbourg), using the comet assay, performed on HeLa S3 cells. In each city, 60-90 non-smoking volunteers composed of two groups of equal size (adults and children) carried the personal Harvard Chempass multi-pollutant sampler during 48h along two different seasons ('hot' and 'cold'). Volunteers were selected so as to live (home and work/school) in 3 different urban sectors contrasted in terms of air pollution within each city (one highly exposed to traffic emissions, one influenced by local industrial sources, and a background urban environment). Genotoxic effects are stronger for PM(2.5) extracts than for PM(10), and greater in winter than in summer. Fine particles collected by subjects living within the traffic proximity sector present the strongest genotoxic responses, especially in the Paris metropolitan area. This work confirms the genotoxic potency of particulate matter (PM(10) and PM(2.5)) organic extracts to which urban populations are exposed.  相似文献

The SEM EDX backscattered electron (BSE) atomic number contrast has been largely used in this work, in combination with conventional secondary electron microanalysis, to investigate the presence of metal particles in airborne particulate collected at three sites (industrial, residential, and rural background) in the Po Valley (Italy). Individual particle x-ray microanalysis was used for this aim. In many cases, the presence of metal particles was not evident by secondary electron imaging and it was instead revealed by BSE detection. Metal particles were observed either as isolated (not clustered to other particles), or gathered together (homogeneous clusters). In addition, the BSE microanalysis put on evidence two main types of association of metals to other particulate components: heterogeneous clusters and metals embedded or enclosed in other materials. In this study, the first association (heterogeneous clusters) was observed mostly between Fe-bearing metallic particles and soot aggregates (or other carbonaceous particles) and it was found in the particulate matter (PM) of all studied sites. The second association, conversely, seems to be characterized by more selective relationships between composition/size of metal particles and type of other particulate components. These associations could be evidenced only when using the BSE Z-contrast and mainly concern three cases: (1) unusual silicate–carbonate mixed aggregates were observed at the industrial site only. In these aggregates, embedded Mn, Cr, Co, Bi, W, and Zr fine particles were selectively observed. (2) Ni and V rich ultrafine particles were only observed as embedded particles in the surface structure of carbon cenospheres. (3) Pb or Pb–Zn bearing fine and ultrafine particles were largely detected only in oxygenated organic aerosols in the ultrafine PM.  相似文献

Background, aim, and scope

Assessment of environmental impacts from pesticide utilization should include genotoxicity studies, where the possible effects of mutagenic/genotoxic substances on individuals are assessed. In this study, the genotoxicity profile of the new formicide Macex® was evaluated with two genotoxicity tests, namely, the micronucleus test with mouse bone marrow and Vicia faba, and a mutagenicity test using the Ames Salmonella assay.

Materials and methods

The bacterial reverse mutation test (Salmonella typhimurium strains TA97, TA98, TA100, TA102, and TA1535), the Vicia root tip and mouse micronucleus tests were conducted according to published protocols.

Results

In the range of the formicide Macex® concentrations tested from 0.06 to 1.0 g?L?1 (or mgkg?1 in the mouse test), no genotoxicity was observed in the prokaryotic or eukaryotic test organisms. However, at Macex® concentrations of 0.5 g?L?1 and above a significant decrease in the mitotic index (P?≤?0.05) in the V. faba was observed. Micronucleus formation was likewise increased in the test organism at concentrations starting at 2.0 g?L?1.

Conclusions

These data allow us to classify this natural formicide preparation as a product with no geno-environmental-impact when applied at recommended concentrations.

In China, the areas that are undergoing rapid urban growth are faced with increasingly more complicated air pollution problems. Sources of air pollution need to be identified and their contributions quantified. In this study, PM2.5 (particulate matter with aerodynamic diameters < or =2.5 microm), PM2.5-10 (particulate matter with aerodynamic diameters 2.5-10 microm), organic carbon (OC), and elemental carbon (EC) concentrations were measured from April to July 2009 at four selected areas in Xiamen (the downtown area, an industrial park, a suburb, and one remote site). The contributions of carbonaceous aerosols to PM2.5 and PM2.5-10 were 20-30% and 10-20%, respectively, indicating that finer particles contained more carbonaceous aerosols. The EC concentrations in PM2.5 at the downtown, industrial, suburb, and remote sites were 2.16 +/- 0.61, 2.05 +/- 0.45, 1.69 +/- 0.54, and 0.65 +/- 0.43 microg m-3, respectively, showing a decrease from the urban and industrial hotspots to the surrounding areas. These data show that carbonaceous aerosols emitted from the combustion of fossil fuels in urban and industrial hotspots influence air quality at the regional scale. Higher levels of PM2.5 and PM2.5-10 were observed at the suburb site compared to the urban and industrial sites. Peak EC concentrations in PM2.5 were observed during the morning and evening rush hours. However, peak PM2.5 levels at the suburb site were observed around noon, which coincides with construction work hours, instead of the morning and evening rush hours when emissions from combustion dominated. These findings indicate that both fuel combustion and construction have exacerbated air pollution in coastal and urban areas in China.  相似文献

Recent developments in scanning electron microscopy (SEM) have produced tabletop instruments capable of reasonable imaging resolution at less cost compared to conventional equipment. Combining the SEM with energy dispersive spectroscopy (EDS) allows the possibility of elemental analysis through detection of X-rays emitted from interaction between individual particles and the SEM electron beam, revealing their atomic composition. It’s well known that exposure to inhalable particulate matter (PM) poses health risks and routine monitoring of the chemical content of these has been realized. Exposure information is of a general character but by combining the chemical build-up of monitored particles and knowledge of their inherent health effects will allow better risk assessment. An analysis technique using a tabletop SEM with EDS is demonstrated on particles collected onto nucleopore filters from urban, industrial and rural areas. Detailed characterization of the instruments analysis capabilities as applied to PM are described.  相似文献

Speciated fine particulate matter (PM2.5) data collected as part of the Speciation Trends Network at four sites in the Midwest (Detroit, MI; Cincinnati, OH; Indianapolis, IN; and Northbrook, IL) and as part of the Interagency Monitoring of Protected Visual Environments program at the rural Bondville, IL, site were analyzed to understand sources contributing to organic carbon (OC) and PM2.5 mass. Positive matrix factorization (PMF) was applied to available data collected from January 2002 through March 2005, and seven to nine factors were identified at each site. Common factors at all of the sites included mobile (gasoline)/secondary organic aerosols with high OC, diesel with a high elemental carbon/OC ratio (only at the urban sites), secondary sulfate, secondary nitrate, soil, and biomass burning. Identified industrial factors included copper smelting (Northbrook, Indianapolis, and Bondville), steel/manufacturing with iron (Northbrook), industrial zinc (Northbrook, Cincinnati, Indianapolis, and Detroit), metal plating with chromium and nickel (Detroit, Indianapolis, and Bondville), mixed industrial with copper and iron (Cincinnati), and limestone with calcium and iron (Bondville). PMF results, on average, accounted for 96% of the measured PM2.5 mass at each site; residuals were consistently within tolerance (+/-3), and goodness-of-fit (Q) was acceptable. Potential source contribution function analysis helped identify regional and local impacts of the identified source types. Secondary sulfate and soil factors showed regional characteristics at each site, whereas industrial sources typically appeared to be locally influenced. These regional factors contributed approximately one third of the total PM2.5 mass, on average, whereas local mobile and industrial sources contributed to the remaining mass. Mobile sources were a major contributor (55-76% at the urban sites) to OC mass, generally with at least twice as much mass from nondiesel sources as from diesel. Regional OC associated with secondary sulfate and soil was generally low.  相似文献

Complex mixtures extracted from air filters exposed for 24 h in two sessions (27 July and 02 August 1991) and at two locations (Merced, downtown, and Pedregal de San Angel, south-west) in Mexico City were analysed. The organic extracts were from airborne particles equal or smaller than 10 microns (PM10), and from total suspended particles (TSP). These organic extracts were assayed in the somatic mutation and recombination test (SMART) in wings of Drosophila melanogaster using two different crosses as well as in the Salmonella/microsome assay using strain TA98 with and without S9 fraction. The presence of polycyclic aromatic hydrocarbons (PAH) in the extracts was determined by gas chromatography. The genotoxic activities observed in the two test systems were comparable with the indirect mutagens producing greater response than the direct mutagens. The quantities of particulate matter as well as the genotoxic activities were higher on 02 August than on 27 July 1991 for both locations. The amounts of airborne particles and the resulting genotoxic activities were higher at Merced than at Pedregal. In both biological systems, PM10 were more genotoxic than TSP. These results demonstrate the sensitivity of the Drosophila wing SMART-which is an in vivo eukaryotic genotoxicity assay-as a biological monitor of environmental pollution related to airborne particles.  相似文献

Approximately 750 total suspended particulates (TSPs) and coarse particulate matter (PM10) filter samples from six urban sites and a background site and >210 source samples were collected in Jiaozuo City during January 2002 to April 2003. They were analyzed for mass and abundances of 25 chemical components. Seven contributive sources were identified, and their contributions to ambient TSP/PM10 levels at the seven sites in three seasons (spring, summer, and winter days) and a "whole" year were estimated by a chemical mass balance (CMB) receptor model. The spatial TSP average was high in spring and winter days at a level of approximately 530 microg/m(3) and low in summer days at 456 microg/m(3); however, the spatial PMo0 average exhibited little variation at a level of approximately 325 microg/m(3), and PM10-to-TSP ratios ranged from 0.58 to 0.81, which suggested heavy particulate matter pollution existing in the urban areas. Apportionment results indicated that geological material was the largest contributor to ambient TSP/PM10 concentrations, followed by dust emissions from construction activities, coal combustion, secondary aerosols, vehicle movement, and other industrial sources. In addition, paved road dust and re-entrained dust were also apportioned to the seven source types and found soil, coal combustion, and construction dust to be the major contributors.  相似文献

Samples of particulate matter were collected during the period from October 2006 through September 2007 in a rural station located 12 km from the city of Colima. A total of 3600 particles were analyzed by SEM-EDS and then classified by their chemical composition and morphology in order of abundance: those rich in C, Fe, Si–Al, Cl–Na, Ca, Ba; and to a lesser abundance, particles rich in heavy metals such as Pb and V–Ni were observed. A factorial analysis was carried out to determine the main elements related with the emission sources such as crustal/farming; fuel-oil; marine; volcanic activity and industry. Trough the seasons, the dominant winds causes the presence of anthropogenic particles in the rural site. The information presented in this study aims to give insight and detailed of the analysis of the morphological characteristics and chemical composition of atmospheric particles at individual level in a rural site from Colima State, Mexico.  相似文献

Plants adapt to metal stress by modifying their metabolism including the production of secondary metabolites in plant tissues. Such changes may impact the diversity and functions of plant associated microbial communities. Our study aimed to evaluate the influence of metals on the secondary metabolism of plants and the indirect impact on rhizosphere bacterial communities. We then compared the secondary metabolites of the hyperaccumulator Pteris vittata L. collected from a contaminated mining site to a non-contaminated site in Vietnam and identified the discriminant metabolites. Our data showed a significant increase in chlorogenic acid derivatives and A-type procyanidin in plant roots at the contaminated site. We hypothesized that the intensive production of these compounds could be part of the antioxidant defense mechanism in response to metals. In parallel, the structure and diversity of bulk soil and rhizosphere communities was studied using high-throughput sequencing. The results showed strong differences in bacterial composition, characterized by the dominance of Proteobacteria and Nitrospira in the contaminated bulk soil, and the enrichment of some potential human pathogens, i.e., Acinetobacter, Mycobacterium, and Cupriavidus in P. vittata’s rhizosphere at the mining site. Overall, metal pollution modified the production of P. vittata secondary metabolites and altered the diversity and structure of bacterial communities. Further investigations are needed to understand whether the plant recruits specific bacteria to adapt to metal stress.  相似文献

Investigations on the monitoring of ambient air levels of atmospheric particulates were developed around a large source of primary anthropogenic particulate emissions: the industrial ceramic area in the province of Castelló (Eastern Spain). Although these primary particulate emissions have a coarse grain-size distribution, the atmospheric transport dominated by the breeze circulation accounts for a grain-size segregation, which results in ambient air particles occurring mainly in the 2.5–10 μm range. The chemical composition of the ceramic particulate emissions is very similar to the crustal end-member but the use of high Al, Ti and Fe as tracer elements as well as a peculiar grain-size distribution in the insoluble major phases allow us to identify the ceramic input in the bulk particulate matter. PM2.5 instead of PM10 monitoring may avoid the interference of crustal particles without a major reduction in the secondary anthropogenic load, with the exception of nitrate. However, a methodology based in PM2.5 measurement alone is not adequate for monitoring the impact of primary particulate emissions (such as ceramic emissions) on air quality, since the major ambient air particles derived from these emissions are mainly in the range of 2.5–10 μm. Consequently, in areas characterised by major secondary particulate emissions, PM2.5 monitoring should detect anthropogenic particulate pollutants without crustal particulate interference, whereas PM10 measurements should be used in areas with major primary anthropogenic particulate emissions.  相似文献

The distribution of mutagenic activity and nitroaromatic components of polycyclic organic matter (POM) in ambient air at industrial, urban, suburban, rural, and remote sites was studied using organic extracts from high volume aerosol samples. Direct-acting mutagens including 1-nitropyrene (1-NP), dinitropyrenes (DNP), and hydroxynitropyrenes (HNP) were measured by high performance liquid chromatography while the mutagenicity was determined in the Salmonella bioassay with strain TA-98. Benzo(a)pyrene (BaP), one of the possible precursors of nitroaromatic compounds in POM, was also measured. In comparing samples from a range of sites, TSP and the concentration of BaP per mass of particulate matter decreased, as expected, at greater distances from urban and industrial combustion sources. However, the concentrations of polar nitroaromatic POM compounds per mass of particles were higher at a remote site than in nonindustrial urban and suburban areas. The mutagenicity in particulate matter extracts from the remote area was predominantly (>90 percent) in the very polar fractions. There were also high atmospheric levels of nitroaromatic compounds and mutagenicity in heavily industrialized areas. These observations may reflect the influences of source emissions, atmospheric transformations of POM compounds, and ther atmospheric processes on the composition of ambient suspended particulate matter.  相似文献

This study investigated water-soluble ions in the sized particles (particularly nano (PM(0.01-0.056))/ultrafine (PM(0.01-0.1))) collected using MOUDI and Nano-MOUDI samplers near a busy road site and at a rural site. The analytical results demonstrate that nano and coarse particles exhibited the highest (16.3%) and lowest (8.37%) nitrate mass ratios, respectively. The mass ratio of NO(3)(-) was higher than that of SO(4)(2-) in all the sized particles at the traffic site. The secondary aerosols all displayed trimodal distributions. The aerosols in ultrafine particles collected at the roadside site exhibited Aitken mode distributions indicating they were of local origin. This finding was not observed for those ultrafine particles collected at the rural site. The mass median diameters (MMDs) of the nano, ultrafine, and fine particles were smaller at the traffic site than at the rural site, possibly related to the contribution of mobile engine emissions.  相似文献

A study of carbonaceous aerosol was initiated in Nanchang, a city in eastern China, for the first time. Daily and diurnal (daytime and nighttime) PM2.5 (particulate matter with aerodynamic diameter < or =2.5 microm) samples were collected at an outdoor site and in three different indoor environments (common office, special printing and copying office, and student dormitory) in a campus of Nanchang University during summer 2009 (5-20 June). Daily PM10 (particulate matter with aerodynamic diameter < or =10 microm) samples were collected only at the outdoor site, whereas PM2.5 samples were collected at both indoor and outdoor sites. Loaded PM2.5 and PM10 samples were analyzed for organic and elemental carbon (OC, EC) by thermal/optical reflectance following the Interagency Monitoring of Protected Visual Environments-Advanced (IMPROVE-A) protocol. Ambient mass concentrations of PM10 and PM2.5 in Nanchang were compared with the air quality standards in China and the United States, and revealed high air pollution levels in Nanchang. PM2.5 accounted for about 70% of PM10, but the ratio of OC and EC in PM2.5 to that in PM10 was higher than 80%, which indicated that OC and EC were mainly distributed in the fine particles. The variations of carbonaceous aerosol between daytime and nighttime indicated that OC was released and formed more rapidly in daytime than in nighttime. OC/EC ratios were used to quantify secondary organic carbon (SOC). The differences in SOC and SOC/OC between daytime and nighttime were useful in interpreting the secondary formation mechanism. The results of (1) OC and EC contributions to PM2.5 at indoor sites and the outdoor site; (2) indoor-outdoor correlation of OC and EC; (3) OC-EC correlation; and (4) relative contributions of indoor and outdoor sources to indoor carbonaceous aerosol indicated that OC indoor sources existed in indoor sites, with the highest OC emissions in I2 (the special printing and copying office), and that indoor EC originated from outdoor sources. The distributions of eight carbon fractions in emissions from the printer and copier showed obviously high OC1 (>20%) and OC2 (approximately 30%), and obviously low EC1-OP (a pyrolyzed carbon fraction) (<10%), when compared with other sources.  相似文献

Atmospheric particulate matter (PM) is an air-suspended mixture of solid and liquid particles that vary in size, shape, and chemical composition. Long-term exposure to elevated concentrations of fine atmospheric particles is considered to pose a health threat to humans and animals. In this context, it has been hypothesized that toxic chemicals such as polycyclic aromatic hydrocarbons (PAHs) play an important role. Some PAHs are known to be carcinogenic and it has been shown that carcinogenic effects of PAHs are mediated by the aryl hydrocarbon receptor (AhR). In this study, PM1 was collected at a rural and an urban traffic site during an intense winter smog period, in which concentration of PM1 often exceeded 50 μg m?3. We applied an in vitro reporter gene assay (DR-CALUX) to detect and quantify PM1-associated chemicals that induce AhR-mediated gene expression. This activity was expressed as CALUX equivalents of 2,3,7,8-tetrachlorodibenzodioxin (PM-TCDD-CEQs). In addition, concentrations of PAHs in the PM1 extracts were determined using gas chromatography/high-resolution mass spectrometry. Concentrations of PM-TCDD-CEQs ranged from 10 to 85 pg m?3 and from 19 to 87 pg m?3 at the urban and rural site, respectively. By the use of known relative potency factors, the measured concentration of a PAH was converted into a PAH-TCDD-CEQ concentration. ΣPAH-TCDD-CEQ and PM-TCDD-CEQ were highly correlated at both sites (r2 = 0.90 and 0.69). The calculated ΣPAH-TCDD-CEQs explain between 2% and 20% of the measured PM-TCDD-CEQs. Benzo[k]fluoranthene was the most important PAH causing approximately 60% of the total ΣPAH-TCDD-CEQ activity. In contrast to NO, CO, PM10, and PM1, the concentration of PM-TCDD-CEQs showed no significant difference between the two sites. No indications were found that road traffic emissions caused elevated concentrations of PM-TCDD-CEQs at the urban traffic site.  相似文献
