Zusammenfassung  Zur ?kotoxikologischen Beurteilung von belastetem Wasser werden h?ufig einfache Biotests eingesetzt. Nicht selten werden dabei toxische Effekte festgestellt. Es stellt sich dann die Frage nach den verantwortlichen Wasserinhalts-stoffen. Durch Festphasenextraktion (SPE) werden diese angereichert, gewonnen und dünnschichtchromatographisch unter Verwendung der automatisierten Mehrfachentwicklung aufgetrennt. Von der DC-Platte wird ein Streifen abgetrennt und darauf direkt die biologische Detektion mit Mikroorganismen (Bacillus subtilis, Leuchtbakterien) durchgeführt. Dadurch k?nnen toxische Banden erkannt werden, Auf dem DC-Plattenrest wird von der toxischen Bande mit einem DC-scanner ein UV-Spektrum aufgenommen, die entsprechende Bande herausgekratzt, der toxische Stoff eluiert und infrarotspektroskopisch untersucht. Durch Spektrenvergleich gelingt es meistens, den Stoff zu charakterisieren. In der vorliegenden Arbeit wird am Beispiel eines Zitzengummieluates das Analysenkonzept vorgestelit. Es zeigte sich, dass dieser Gummiartikel einen Vulkanisationsbeschleuniger (2-Mercaptobenzothiazol) freisetzt. Online-Publikation am: 21.12.1999  相似文献

Use and interaction of navigation strategies in regionalized environments   总被引:2,自引:0,他引:2  
In this work, three experiments are reported that studied the use and interaction of navigation strategies both during the learning of a virtual environment and during subsequent route planning tasks. Special interest concerned the role of regions within the environments. Results from Experiment 1 suggest that the regions are perceived and encoded in spatial memory very early during the process of learning an environment. During navigation such regional information could be used to overcome missing or imprecise spatial information on the detailed level. Experiments 2 and 3 studied the use and interaction of route planning strategies that are applied after an environment has been learned. Results suggest (i) that human route planning takes into account region-connectivity and is not based on place-connectivity alone, (ii) that route planning takes into account the distribution of multiple target locations and (iii) that route planning takes into account the complexity of alternative paths.  相似文献

Atmospheric particulate matter (PM) is an air-suspended mixture of solid and liquid particles that vary in size, shape, and chemical composition. Long-term exposure to elevated concentrations of fine atmospheric particles is considered to pose a health threat to humans and animals. In this context, it has been hypothesized that toxic chemicals such as polycyclic aromatic hydrocarbons (PAHs) play an important role. Some PAHs are known to be carcinogenic and it has been shown that carcinogenic effects of PAHs are mediated by the aryl hydrocarbon receptor (AhR). In this study, PM1 was collected at a rural and an urban traffic site during an intense winter smog period, in which concentration of PM1 often exceeded 50 μg m?3. We applied an in vitro reporter gene assay (DR-CALUX) to detect and quantify PM1-associated chemicals that induce AhR-mediated gene expression. This activity was expressed as CALUX equivalents of 2,3,7,8-tetrachlorodibenzodioxin (PM-TCDD-CEQs). In addition, concentrations of PAHs in the PM1 extracts were determined using gas chromatography/high-resolution mass spectrometry. Concentrations of PM-TCDD-CEQs ranged from 10 to 85 pg m?3 and from 19 to 87 pg m?3 at the urban and rural site, respectively. By the use of known relative potency factors, the measured concentration of a PAH was converted into a PAH-TCDD-CEQ concentration. ΣPAH-TCDD-CEQ and PM-TCDD-CEQ were highly correlated at both sites (r2 = 0.90 and 0.69). The calculated ΣPAH-TCDD-CEQs explain between 2% and 20% of the measured PM-TCDD-CEQs. Benzo[k]fluoranthene was the most important PAH causing approximately 60% of the total ΣPAH-TCDD-CEQ activity. In contrast to NO, CO, PM10, and PM1, the concentration of PM-TCDD-CEQs showed no significant difference between the two sites. No indications were found that road traffic emissions caused elevated concentrations of PM-TCDD-CEQs at the urban traffic site.  相似文献

Amounts of readily soluble nutrients on asphalt parking lot surfaceswere measured at four locations in metropolitan Phoenix, Arizona, U.S.A. Using a rainfall simulator, short intense rainfall events were generated to simulate `first flush' runoff. Samples were collected from 0.3 m2 sections of asphalt at 8 to 10 sites on each of four parkinglots, during the pre-monsoon season in June-July 1998 and analyzed for dissolved NO3 --N, NH4 +-N, soluble reactive phosphate (SRP), and dissolved organic carbon (DOC). Runoff concentrations varied considerably for NO3 --N and NH4 +-N (between 0.1 and 115.8 mg L-1) and DOC (26.1 to 295.7 mg L-1), but less so for SRP (0.1 to 1.0 mg L-1), representing average surface loadings of 191.3, 532.2, and 1.8 mg m-2 respectively. Compared with similar data collected from undeveloped desert soil surfaces outside the city, loadings of NO3 --N and NH4 +-N on asphalt surfaces were greater by factors of 91 and 13, respectively. In contrast, SRP loads showed little difference between asphalt and desert surfaces. Nutrient fluxes in runoff from a storm that occurred shortly after the experiments were used to estimate input-output budgets for 3 of the lots under study. Measured outputs of DOC and SRP were similar to those predicted using rainfall and experimentally determined surface loadings, but for NH4 +-N and particularly for NO3 --N, estimated rainfall inputs and surface runoff were significantly higher than exports in runoff. This suggests that parking lots may be important sites for nutrient accumulation and temporary storage in arid urban catchments.  相似文献

Polychlorinated biphenyls (PCBs) and the structurally related polybrominated diphenyl ethers (PBDEs) have been associated with chronic neurotoxicity involving reduced motor activity and impaired attentiveness. Such neurobehavioral effects indicate that the central nervous system may represent an important target organ for the action of these persistent contaminants in wildlife. As a consequence, the brain of different terrestrial and aquatic birds collected in Switzerland was analysed for PCBs and PBDEs. In parallel, the same contaminants were examined in the accompanying adipose tissue. After clean-up by means of glass columns containing acidified silica, deactivated alumina and anhydrous sodium sulphate, the samples were analysed by high resolution gas chromatography/tandem mass spectrometry (HRGC-MS/MS). Median PCB concentrations in the brain (sum of PCB 28, PCB 52, PCB 101, PCB 118, PCB 138, PCB 153 and PCB 180) ranged between 13 ng g(-1) wet weight (ww) in blackbirds (Turdus merula) and 428 ng g(-1) ww in sparrow hawks (Accipiter nisus). Median PBDE concentrations in the brain (sum of BDE 28, BDE 47, BDE 99, BDE 100, BDE 153, BDE 154 and BDE 183) ranged from below the decision limit in buzzards (Buteo buteo) and blackbirds, to 14 ng g(-1) ww in sparrow hawks. After correction for the respective lipid content, higher PCB or PBDE concentrations in brain compared to adipose tissue, were found in three sparrow hawks, four buzzards and in all investigated blackbirds. These results suggest that a deficit in the neuroprotective function of the blood-brain barrier may cause unexpected levels of PCBs and PBDEs in the central nervous system.  相似文献

Simple biotests are frequently used for the ecotoxicological assessment of waste water. Unfortunately, and not too rarely, effects are detected and the question concerning water-polluting substances has to be answered. The latter are enriched by solid phase extraction (SPE), then isolated and fractioned by thinlayer chromatography using automatic multiple development (AMD). One strip of the DC-plate is separated and used without further atterations for the biological detection with microorganisms (Bacillus subtilis, luminous bacteria). With this system, toxic bands can be identified. From the other part of the DC plate, an UV-spectrum can be taken of the toxic bands using a DC scanner. Subsequently, the corresponding bands are scratched out and the toxic substance is eluted and analysed with infrared spectroscopy. The infrared spectrum can be characterised by comparing it with reference spectra. In this submitted work, a concept is presented with an example of a “nipple rubber” eluate. It shows that this rubber article liberates a vulcanization accelerator, 2-mercaptobenzothiazole.  相似文献
