The effects of 32 pesticides at two concentrations on acetylene reduction (non-symbiotic nitrogen fixation), nitrogen fixers, bacteria and fungi in an organic soil were assessed. None of the pesticide treatments suppressed C2H2 reduction as compared to controls. No significant inhibition of the population of non-symbiotic nitrogen fixers occurred. However, stimulatory effects were observed with treatments of fensulfothion, fonofos, oxamyl, DDR, TeloneR and Telone CR. Bacterial and fungal populations showed temporary declines but all recovered within 7 days to levels similar to or higher than those in the controls.  相似文献

Abstract

The objective of this study was to determine the effects of varying nitrogen sources and concentrations upon glutamine synthetase and protease activities in Prevotella ruminicola strain B14. Based on growth response it appears that ammonium chloride or pepticase limited P. ruminicola becomes nitrogen limited when nitrogen concentration is at 0.5 mM. However, when casein was provided as the sole source of nitrogen P. ruminicola becomes nitrogen limited at 2.5 mM. Glutamine synthetase activity was measured from mid‐log phase cells grown in either nitrogen‐limited or non‐limited conditions. No activity was detectable in the non‐limited treatments. However, in the N‐ limited treatments, pepticase had the highest activity (20.76 units), followed by ammonium chloride (18.72 units) and casein (14.42 units). Protease activity assays indicated that nitrogen‐limited cultures had higher proteolytic activity than non‐limited cultures. Moreover, these activities appeared to follow the same response pattern as the previously observed glutamine synthetase activities. The results of this study indicate that P. ruminicola strain B, 4 protease activity may be influenced by nitrogen concentration such that activity increases when nitrogen availability decreases.  相似文献

Abstract

Levels of acephate (OrtheneR) and its principle metabolite, methamidophos, in/on greenhouse‐grown pepper and cucumber fruits and leaves in relation to the applied methamidophos were monitored. Dislodgeable and total residues of acephate and methamidophos were determined by gas‐liquid chromatography equipped with a flame ionization detector (GC‐FID) and were confirmed by nitrogen phosphorus detector (GC‐NPD). The dissipation curves of the residues followed first‐order kinetics (R2> 0.96). Initial residues of acephate on fruits varied between pepper (15.12 ppm) and cucumber (2.16 ppm) . Total residues in fruits and leaves determined at intervals following application revealed the greater persistence of acephate on pepper fruits (half‐life [t1/2] of 6 d) than on cucumber fruits (t1/2 was 3.7 d) . T1/2 values for the applied methamidophos were 4.7 and 5.3 d on pepper and cucumber fruits, respectively. Deacety‐lation of acephate (formation of its metabolite) was detectable 1 d following acephate treatment and reached a maximum of 2.05% of initial acephate residues 3 d after application on pepper fruits. On cucumber fruits, acephate metabolite reached a maximum of 2.12% one wk following application. No acephate residues were detected above the limit of detection of 0.001 ppm in pepper fruits 50 d following acephate application while its metabolite was detectable at that time (detectability limit was 0.0001 ppm).  相似文献

水葫芦对滇池底泥氮磷营养盐释放的影响   总被引:1,自引:0,他引:1  
为了探讨水葫芦对富营养湖泊底泥氮磷营养盐释放的影响,通过原位采集滇池柱状底泥,以蒸馏水为上覆水,进行了25d的室内静态模拟实验。实验比较了水葫芦处理组和空白对照组底泥氨氮(NH4-N)、硝态氮(NO3^-N)、溶解性总氮(DTN)、正磷酸盐(PO4^3--P)等的释放特征。结果表明,与对照组相比,水葫芦处理组上覆水溶解氧、pH显著性降低,而PO4^3-P浓度显著性升高;在实验前2d,水葫芦处理组上覆水NH4^+N和DTN浓度显著性高于对照组,而在5~25d时,其显著性低于对照组。根据上覆水营养盐浓度、水葫芦植株吸收营养盐总量,推算底泥氮磷营养盐释放的平均速率,表明水葫芦加速了滇池底泥氮、磷营养盐的释放速率,处理组氮、磷释放速率分别为对照组的5.3~170.2和1.5~21.6倍。  相似文献

Abstract

The immuno‐biochemical effects of cobaltous chloride in rats receiving iron‐sufficient and deficient diets were investigated. Rats receiving 100 ppm or more cobalt showed a significant reduction in thymus and body weights along with a marked decrease in hemoglobin, hematocrit, sheep agglutinins and plaque forming cells. These effects were more pronounced in rats receiving cobalt mixed with iron‐deficient diet than those fed on iron‐sufficient diet. The Na+‐K+ and mitochondrial (Oligomycin‐sensitive) Mg2+ATPase activities in brain and liver of rats fed with iron‐deficient diets were decreased significantly. However, the ATPase activities in these tissues from rats fed with cobalt mixed with iron‐sufficient diets were not altered.  相似文献

Ammonia nitrogen pollution control is an urgent issue of landfill. This research aims to select an optimal refuse for ammonia nitrogen removal in landfill from the point of view of adsorption and desorption behavior. MSW (municipal solid waste) samples which deposit ages were in the range of 5 to 15 years (named as R15, R11, R7, and R5) were collected from real landfill site. The ammonia nitrogen adsorption behaviors of MSW including equilibrium time, adsorption isotherms, and desorption behaviors including equilibrium time were determined. Furthermore, the effects of pH, OM, Cu(II), Zn(II), and Pb(II) on adsorption and desorption behavior of ammonia nitrogen were conducted by orthogonal experiment. The equilibrium time of ammonia nitrogen adsorption by each tested MSW was very short, i.e., 20 min, whereas desorption process needed 24 h and the ammonia nitrogen released from refuses was much lesser than that adsorbed, i.e., accounted for 3.20 % (R15), 14.32 % (R11), 20.59 % (R7), and 20.50 % (R5) of each adsorption quantity, respectively. The maximum adsorption capacity estimated from Langmuir isotherm appeared in R15-KCl, i.e., 25,000 mg kg?1. The best condition for ammonia nitrogen removal from leachate was pH >7.5, OM 23.58 %, Cu(II) <5 mg L?1, Zn(II) <10 mg L?1, and Pb(II) <1 mg L?1. Ammonia nitrogen in landfill leachate could be quickly and largely absorbed by MSW but slowly and infrequently released. The refuse deposited for 15 years could be a suitable material for ammonia nitrogen removal.  相似文献

Abstract

The effects of temperatures and solar radiation on the dissipation of 14C‐p,p'‐DDT from a loam soil was studied by quantifying volatilization, mineralization and binding. The major DDT loss occurred by volatilization, which was 1.8 times more at 45oC than at ambient temperature (30°C). Mineralization of DDT slowly increased with time but it decreased slightly with increase in temperature. Binding of DDT to soil was found to be less at higher temperatures (35 and 45°C) as compared to ambient temperature. Degradation of DDT to DDE was faster at higher temperatures.

Exposure of non‐sterilized and sterilized soils treated with 14C‐DDT to sunlight in quartz and dark tubes for 6 weeks resulted in significant losses. Volatilization and mineralization in quartz tubes were more as compared to dark tubes. The volatilized organics from the quartz tubes contained larger amounts of p,p'‐DDE than the dark tubes. Further, higher rates of volatilization were found in non‐sterilized soils than in sterilized soils. The results suggest that faster dissipation of DDT from soil under local conditions relates predominantly to increased volatilization as influenced by high temperature and intense solar radiation.  相似文献

Abstract

A laboratory study was conducted to examine the effects of five insecticides on microbial and enzymatic activities important to fertility in sandy soil. Cyfluthrin significantly increased bacterial populations after 2 wks. Imidacloprid showed an inhibitory effect on fungal numbers after 2 wks incubation while the others did not affect fungal population. No inhibitory effect was observed on nitrification of soil indigenous nitrogen. All treatments stimulated S‐oxidation after 4 wks. With the exception of cyfluthrin and imidacloprid after 2 wks, denitrification in sandy soil indicated that all treatment inhibited denitrification throughout the experiment. No inhibitory effects on biomass‐c were observed during 2‐wk periods. An inhibitory effect was observed on amylase after 1 wk while significant recovery was observed after 3 wks. With the exception of HgCl2, no effect was observed on reducing sugar formation for 2 wks with all treatments. Formazan formation resulting from dehydrogenase activity was significantly greater with tebupirimphos and Aztec for 1 wk. All treatments supressed phosphatase activity for 1 wk, while none of the treatments suppressed phosphatase activity after 2 wks. Amitraz, tebupirimphos and Aztec inhibited urease activity for 1 wk. With the exception of tebupirimphos, no treatments affected N2‐fixation in soil. Although short‐lived inhibitory effects on activities of microbes and enzymes were caused by the insecticides, the soil indigenous microbes can tolerate the chemicals used for control of soil pests.  相似文献

This study investigated potential nitrogen fixation, net nitrification, and denitrification responses to short-term crude oil exposure that simulated oil exposure in Juncus roemerianus salt marsh sediments previously impacted following the Deepwater Horizon accident. Temperature as well as crude oil amount and type affected the nitrogen cycling rates. Total nitrogen fixation rates increased 44 and 194 % at 30 °C in 4,000 mg kg?1 tar ball and 10,000 mg kg?1 moderately weathered crude oil treatments, respectively; however, there was no difference from the controls at 10 and 20 °C. Net nitrification rates showed production at 20 °C and consumption at 10 and 30 °C in all oil treatments and controls. Potential denitrification rates were higher than controls in the 10 and 30?ºC treatments but responded differently to the oil type and amount. The highest rates of potential denitrification (12.7?±?1.0 nmol N g?1 wet h?1) were observed in the highly weathered 4,000 mg kg?1 oil treatment at 30 °C, suggesting increased rates of denitrification during the warmer summer months. These results indicate that the impacts on nitrogen cycling from a recurring oil spill could depend on the time of the year as well as the amount and type of oil contaminating the marsh. The study provides evidence for impact on nitrogen cycling in coastal marshes that are vulnerable to repeated hydrocarbon exposure.  相似文献

Abstract

The effects of temperature and solar radiation on dissipation of 14C‐p,p'‐DDT from a latosol soil were studied under laboratory conditions. Volatilization was measured by trapping organic volatiles during 6 weeks and was found to increase with rise of temperature from 3.8% of initial amount at ambient temperature to 5.9% at 45°C.

Studies on the effect of solar radiation using quartz tubes under sterilized and non‐sterilized conditions have shown that volatilized organics were highest in quartz tubes, with soil microflora presumably playing a very minor role in volatilization. Mineralization was shown to be low in sterilized systems and highest in non‐sterilized quartz systems. Studies on binding suggest that soil bioactivity may be involved in the formation of a portion of the bound residue. These laboratory experiments seem to support data from the field, where it is maintained that volatilization is a major mechanism for dissipation. Degradation in soil and to a lesser extent solar irradiation contribute also substantially to the dissipation mechanisms. Radiocarbon dissipated from plywood surfaces under indoor conditions in a biphasic fashion. Loss of 50% occurred after 5.5 weeks while the remainder dissipated at a very slow rate.  相似文献

Abstract

Penicillic acid and ochratoxin A are environmentally important toxic fungal metabolites (mycotoxins) that are synergistic in combination. The effects of penicillic acid on the pancreatic enzyme, :arboxypeptidase A were investigated in vitro and in vivo. A broad range of inhibition in vitro of the enzyme by PA was demonstrated with a half‐maximal inhibitory concentration equal to 1.1 x 10‐4M PA. Inhibition of carboxypeptidase A was time and temperature dependent, and resulted in decreased conversion of parent ochratoxin A to the non‐toxic metabolite, ochratoxin alpha. Studies in vivo demonstrated a penicillic acid‐dependent inhibition of pancreatic carboxypeptidase A activity in the mouse and the chicken following multiple oral exposure. It is postulated that the mode of toxic interaction of the two mycotoxins may be due, in part, to impaired detoxification of ochratoxin A through peni‐cillic acid depletion of carboxypeptidase A activity.  相似文献

Abstract

Laboratory tests were conducted with 14 insecticides applied to a sandy loam to determine whether these materials caused any effects on microbial activities related to soil fertility. The ammonification of soil native organic nitrogen was not inhibited by any of the insecticide treatments. Some insecticides showed an effect on nitrification during the second week of incubation which subsequently recovered to levels similar to those obtained in the controls. There was a significant effect on denitrification in a number of treated samples. However, recovery of denitrifying capacity was rapid. This recovery indicated that the indigenous soil microorganisms can tolerate the chemicals used for control of insect pests. No significant inhibition of sulfur oxidation was observed. Results indicated that the insecticide treatments at the level tested were not drastic enough to be considered deleterious to soil microbial activities important to soil fertility.  相似文献

Abstract

Potatoes were grown from cut seed in Plainfield sand treated in‐furrow with disulfoton (Di‐Syston 15G, 3.36 kg Al/ha) in 1983 and from whole seed in similarly treated loam in 1991. Soils were contained in 2 m2 field plots. Soil, seed potato and foliage were analyzed for the insecticide and its sulfoxide and sulfone metabolites during the 8–12 wk following planting. Disulfoton disappeared at different rates from the two soils (ksand=0.024 day‐1, kloam=0.056 day‐1) with partial conversion to the sulfoxide and sulfone in both. Larger quantities of the three insecticidal components were absorbed by the seed potato in the cut‐seed/sand combination. The relative amounts of these components in the seed potato also differed between treatments with disulfoton being the largest component of the cut‐seed/sand and smallest in the whole‐seed/loam. Disulfoton sulfoxide and sulfone were the major insecticidal components of the foliage and concentrations in the initial foliage (each ca. 10 ppm) were similar for both treatments. Sulfoxide concentrations in the foliage decreased more rapidly than the sulfone and the decrease in concentration of each of the components was similar for the two treatments.  相似文献

Abstract

The sorption of imidacloprid (l‐[(6‐chloro‐3‐pyridinyl)‐methyl]‐N‐nitro‐2‐imidazolid‐inimine) (IMI) and its metabolites imidacloprid‐urea (l‐[(6‐chloro‐3‐pyridinyl)‐methyl]‐2‐imidazol‐idinone) (IU), imidacloprid‐guanidine (l‐[(6‐chloro‐3‐pyridinyl)‐methyl]‐4,5‐dihydro‐lH‐imidazol‐2‐amine) (IG), and imida‐cloprid‐guanidine‐olefin ( 1 ‐[(6‐chloro‐3‐pyridinyl)methyl]‐lH‐imidazol‐2‐amine) (IGO) was determined on six typical Brazilian soils. Sorption of the chemicals on the soil was characterized using the batch equilibration method. The range and order of sorption (Kd) on the six soils was IG (4.75–134) > IGO (2.87–72.3) > IMI (0.55 ‐16.9) > IU (0.31–9.50). For IMI and IU, Kd was correlated with soil organic carbon (OC) content and CEC, the latter due to the high correlation between OC and cation exchange capacity (CEC) (R2=0.98). For IG and IGO, there was no correlation of sorption to clay, pH, OC or CEC due to the high sorption on all soils. Average Koc values were IU = 170, IMI = 362, IGO = 2433, and IG = 3500. Although Kd and Koc values found were consistently lower than those found in soils developed in non‐tropical climates, imidacloprid and its metabolites were still considered to be slightly mobile to immobile in Brazilian soils.  相似文献

Abstract

The effect of intratracheal administration of DDT (5 mg/100 g body weight) or endosulfan (1 mg/100 g body weight) to rats for three consecutive days, has been studied on liver lipid metabolism. The administration of DDT but not endosulfan significantly increased the liver weight and the microsomal protein contents. Both DDT and endosulfan treatments significantly increased the contents of microsomal phosphatidylcholine (PC), total‐free‐ and esterified cholesterol. The distribution of unsaturated fatty acids of microsomal PC and PE was increased by DDT treatment. The intratracheal administration of DDT caused fatty infiltration of liver which was probably due to increased synthesis of triglycerides (TG). This is supported by the increased incorporation of radioactive palmitate‐l‐14C into microsomal TG. However, the increased incorporation of palmitate‐l‐14C into microsomal PC and phosphatidylethanolamine (PE) after the DDT treatment, was due to the increased transacylation reaction supported by the decreased activity of microsomal phospholipase A. The intratracheal adminstration of endosulfan did not has pronounced effect on liver fatty infiltration, or transacylation reaction in microsomal PC and PE. However, the results have shown that the treatments of DDT or endosulfan increased the PC contents and the incorporation of radioactive [methyl‐3H]choline into PC of microsomes, resulting the increased synthesis of PC via CDPcholine pathway. Thus, the intratracheally administered DDT or endosulfan to rats showed that both the insecticides cause manifestations in the biochemistry of microsomal membrane lipids, although the effects of DDT being more pronounced. Therefore, the translocation effects of these insecticides or metabolites from lung to liver is established.  相似文献

To elucidate the roles of microorganisms in the nitrogen geo-chemical cycle, the distributions of nitrogen-utilising bacteria were studied in a small eutrophic lake. Ammonifiers, nitrogen fixers and nitrate reducers were more concentrated at the severely polluted Xili sub-area than at the Centre of Outer (CO), whereas the contrary was true for autolithotrophic nitrifiers. Molecular identification of predominant ammonifiers and nitrogen fixers showed that two of the ammonifiers were γ-Proteobacteria, two were β-Proteobacteria, one was CFB group and the other three were low G+C% Gram positive Bacillus. Among four nitrogen fixers, one belonged to α-Proteobacteria, and the other three were γ-Proteobacteria.  相似文献

Abstract

The immunosuppressive effect of lindane was assessed in weanling rats over a period of 5 weeks. Agglutinin titres against S. typhi ‘O’ and ‘H’, S. paratyphi ‘AH’ and ‘BR’ antigens were determined following TAB vaccination in control and lindane‐fed animals. The titres attained in non‐treated controls were higher significantly than those in treated animals indicating a suppressive effect of lindane on humoral immune responses.  相似文献

Abstract

A study was undertaken to compare and evaluate the different types of 63Ni and 3H electron capture (ECD) and flame photometric detectors (FPD) as part of the Canadian Check Sample Program on Pesticide Residue Analysis. Twenty‐seven laboratories were supplied with chlorpyrifos standard solutions for the determination of linear range and minimum detectable amounts (MDA). Each laboratory selected its own operating conditions except for the FPD study. Results for the FPD were compared with standardized flame conditions designed to optimize the exponential factor in the S‐mode. The MDA of the ECDs (pulsed and direct‐current) ranged from 0.09 to 0.17 pg/sec for 63Ni and 3H sources respectively. The corresponding MDA for the linearized ECDs (pulse modulated or constant current) was 0.04 pg/sec and a linear range of 104 or greater with nitrogen as the carried gas. Use of argon/methane and a pulse width of 0.1 usec extended this range to 105 but the sensitivity was not as great as with nitrogen carrier gas. The practical limit of detector sensitivity was found to be instrument dependent depending primarily on the noise level. In the FPD study, operating in the P‐mode gave an average MDA of 5.97 pg/sec and a linear range of 2 x 103. Similarly, the S‐mode parameters were 81.1 pg/sec and 8 x 102 respectively with an average exponential factor of n 1.8. Attempts to apply optimized detector gas‐flow conditions improved sensitivity and linearity in the S‐mode only.  相似文献

Abstract

The effects of low levels of diazinon treatment on four marker enzymes in rat heart and skeletal muscle have been investigated. Adult male Wistar rats were treated twice a week with a dose of 0.5 ml‐kg‐1day‐1 diazinon for 28 weeks. Diazinon treated rats gained significantly less weight than Sham‐treated controls. Typical differences in Succinate dehydrogenase (SDH), Lactate dehydrogenase (LDH), Phosphofructo kinase (PFK) and Hexokinase (HK) activities were observed between heart and skeletal muscles. Diazinon feeding had no effect on heart, soleus, gastrocnemius and plantaris SDH, LDH and PFK enzyme activities after 28 weeks. HK activity was significantly increased in sham‐control soieus and plantaris muscles after 28 weeks. Diazinon feeding inhibited HK activity in plantaris muscle after 28 weeks treatment These results demonstrate that chronic low levels of diazinon have little effect on the glycolytic and oxidative activity in heart and skeletal muscle.  相似文献

Abstract

The degradation of [phenyl‐U‐14C]methabenzthiazuron (MBT) and formation of bound residues in the surface soil of an orthic luvisol were studied under constant climatic conditions (20°C, 40 % of maximum water holding capacity). In two treatments (with and without preincubation in the soil) maize straw was amended at a rate of 1.5 g/100 g dry soil in addition to the application of MBT. The mineralization of uniformly labeled maize straw was studied simultaneously. In additional flasks, MBT was incubated at 0, 10 and 30°C with and without addition of maize straw.

The turnover of the amended maize straw led to an enhanced dissipation of MBT which was mainly due to the formation of bound residues. This corresponded to a higher microbial activity in the soil after straw amendment and the intensive mineralization of the radiolabeled maize straw. About 2–3 % of the applied radioactivity from the radiolabeled maize straw was measured in the soil microbial biomass 10 and 40 days after application whereas 14C from MBT was only incorporated into soil microbial biomass in the treatments with straw amendment.

Within the bound residue fractions relatively more radioactivity was measured in fulvic and humic acids after straw amendment. Increasing temperatures promoted the dissipation of MBT and the formation of bound residues in both treatments, but without amendment of maize straw these effects were far less pronounced. The laboratory scale degradation experiment led to similar results as were found in a corresponding lysimeter study. Differences that were observed could be explained by different temperature regimes of the experiments and time of aging in soil.  相似文献
