近海养虾场底泥中产芽孢细菌的生态特征   总被引:7,自引:0,他引:7  
通过对近海养虾场底泥中的细菌数量和类群的调查,发现有超过50%的细菌生物量是产芽孢细菌,因此对底泥中的产芽孢细菌进行了分离和纯化,通过对细胞形态、生理生化等特征的研究和对部分菌株的16S rRNA基因的ARDRA分型、序列分析等,鉴定了67株产芽孢细菌,其中62株属于芽孢杆菌属,5株属于短芽孢杆菌属.进一步对62株芽孢杆菌属的细菌在底泥不同深度的分布进行研究,结果表明,巨大芽孢杆菌主要分布在底泥深度0~6cm左右的区域,海洋芽孢杆菌、短小芽孢杆菌和蜡状芽孢杆菌主要分布于底泥6cm以下的区域,与坚强芽孢杆菌性状相近的菌分布在底泥2~8cm深度;与耐碱芽孢杆菌性状相近的芽孢菌广泛分布在0~12cm区域.讨论认为,应用这些产芽孢细菌资源在修复海洋环境和开发海水养殖微生态制剂方面具有一定可能性.图3表3参15  相似文献

为了给饮用水低温生物预处理技术的应用与开发提供实验依据,利用陶粒柱连续流生物反应器,于低温(水温2─5℃)季节,对某水源水进行预处理的同时,研究和探讨了陶粒柱生物反应器的生化活性。结果表明,试验装置在低温季节对原水的净化效果较好;陶粒柱反应器内TTC-脱氢酶活性沿水流方向自上而下呈梯度分布;低温条件下测定的反应器上、中、下层生物陶粒TTC-脱氢酶活性分别为22.39、5.70、0.408μgTF/cm3·h-1;生物陶粒TTC-脱氢酶活性与活性细菌数的时数(logABN)之间具有显著相关性(r=0.8493.n=25)。因此,可以认为,TTC-脱氢酶活性是研究和描述饮用水生物预处理效果的有效参数。  相似文献

以国内外不同品种、不同陈化时间的10个烟叶样品为材料,对其叶面高温微生物进行了分离纯化、鉴定和多样性研究,以了解表面高温微生物在烟叶陈化发酵中的作用.结果表明:陈化烟叶表面高温菌总量为103~105个/g,高温细菌占绝对优势,真菌很少,未能分离到放线菌和酵母菌;高温细菌主要包括芽孢杆菌属(Bacillus)和地芽孢杆菌属(Geobacillus)两个属,芽孢杆菌属为优势菌属;高温霉菌主要包括青霉属(Penicillium)和曲霉属(Aspergillus),曲霉属为优势菌属.国内外烟叶表面微生物种类和数量有所差异,与陈化时间和烟叶品种等因素有着密切关系.  相似文献

通过分析农田、林地、草地和菜地土壤有机磷细菌和无机磷细菌的数量及种群结构,发现有机磷细菌数量比无机磷细菌多;有机磷细菌主要是芽孢杆菌属,其次是假单胞菌属;而无机磷细菌主要是假单胞菌属。菜地土壤解磷细菌的数量和种类最多。不同的细菌解磷能力差异很大,其解磷能力与培养介质的pH值有一定的关系;许多菌株在进一步的纯化过程中失去了解磷能力。  相似文献

白芨滩国家级自然保护区及其周边植被恢复有利于改善区域生态环境,而植被恢复良好的区域内镶嵌分布着大面积藓结皮和藻结皮,其在防风固沙、改善荒漠生态系统地表土壤质量等方面发挥着重要作用。为深入了解该区域不同生物土壤结皮中微生物群落结构及其差异,选取藓结皮和藻结皮镶嵌分布的地表覆盖区设置取样点,分别对藓结皮和藻结皮进行细菌和真菌的Illumina MiSeq测序。结果表明,不同生物土壤结皮中细菌群落和真菌群落α多样性趋于一致,仅反映细菌群落丰富度的ACE和Chao指数在藓结皮中显著高于藻结皮。不同生物土壤结皮形成以微枝形杆菌属、红色杆菌属、地嗜皮菌属、芽球菌属、鞘氨醇单胞菌属、土壤红杆菌属、微红微球菌属、类诺卡氏菌属和苔藓杆菌属等为主要类群的细菌群落结构以及以赤霉菌属、Knufia、被孢霉属、石果衣属、链格孢属、Bahusakala、Phaeomycocentrospora、光黑壳属和Omphalina等属为主要已知类群的真菌群落结构。藓结皮中,红色杆菌属细菌相对丰度显著低于藻结皮,而苔藓杆菌属细菌相对丰度显著高于藻结皮。藓结皮中,Knufia、Bahusakala、Omphalina、Lam...  相似文献

从地质微生物生态学的角度研究和阐述了云南和西藏热泉中主要微生物类群及其金成矿作用.发现了热泉中存在的嗜热好氧菌优势菌属为并孢杆菌(Racillus),放线菌中的优势菌属为链霉菌(Streptomyces),真菌中优势菌属为曲霉(Aspergillus),厌氧菌中优势菌为硫酸盐还原菌的脱硫肠状菌属(Desulfotomaculum)和核状芽孢杆菌属(Clostridium),对吸附聚金、沉淀金的功能菌进行的研究发现.嗜热细菌对溶解金有明显的吸附聚金作用,表明原溶液金大多富集于细胞体;并以芽孢杆菌属K12-1菌株等进行了吸附聚金条件试验.  相似文献

利用气体循环培养体系从陕西乾县HUP-豆科植物紫花苜蓿(Medicago sativa)根际土壤中分离获得37株细菌.菌株氧化氢能力测定结果表明,8株菌氧化氢和自养生长能力较强,初步确定为氢氧化细菌类群;根据其形态特征、培养特征和生理生化特性,鉴定为7个不同属:假单胞菌属(Pseudomonas)、邻单胞菌属(Plesiomonas)、脂肪杆菌属(Pimelobacter)、黄色杆菌属(Xanthobacter)、勒米诺氏菌属(Leminorella)、地杆菌属(Terrabacter)和稀有杆菌属(Rarobacter);其中氧化氢能力最强的优势菌株WMQ-7 16S rDNA序列(GenBank登录号为EU807744)长度为1 451bp,GC含量为53.8%,其核苷酸序列与假单胞菌属同源性高于99%,在系统发育树上位于同一分支,将WMQ-7菌株鉴定为假单胞菌属(Pseudomonas).图2表5参16  相似文献

对松嫩平原苏打盐碱地养鱼稻田水体微生物数量及种类组成进行了初步研究。结果表明,养鱼稻田异养细菌数量为0.280万~30.716万mL-1,高于未养鱼稻田(P<0.05),其平均值的时间分布为水稻生长末期>中期>初期;养鱼稻田和未养鱼稻田放线菌和霉菌数量均较少。养鱼稻田大肠菌群数量为403.6~17934.2L-1,显著高于未养鱼稻田(P<0.01),其平均值的时间分布为水稻生长中期>初期>末期。异养细菌数量与鱼产量、总施肥量和有机肥施用量均呈极显著相关,与化肥施用量相关不显著;大肠菌群数量与鱼产量相关显著,与总施肥量相关不显著,但与有机肥施用量相关极显著。养鱼稻田中检测出10个科(属)异养细菌,优势菌为弧菌属(Vib rion)、气单胞菌属(Aeromonas)和芽孢杆菌属(Bacillus)。  相似文献

从新疆、内蒙干旱地区苜蓿属、草木樨属、锦鸡儿属植物的根瘤中分离出54株根瘤菌,对其中48株根瘤菌和22株参比菌一起进行了数值分类研究,在85%的相似性水平上,未知菌分为3个不同于已知种的新群.另加入6株分离自锦鸡儿属植物的根瘤菌,共76株菌进行了全细胞蛋白SDSPAGE分析,在80%的相似性水平上,已知菌相应成群,未知菌除XJ96342在68%的相似性水平上和群5、6、7、8、R.galega聚群外,其余未知菌则分布在4、5、6、7、8、9、10七个群中.群4中所有菌株属于数值分类的群1;群6中有3株菌属于数值分类的群3;群9中有8株菌为数值分类群2的菌株.反映了两种方法在分类上得到的结果比较近似.  相似文献

菜心(Brassica parachinensis)是华南地区的一种特色蔬菜.为探究无土栽培和土壤栽培菜心内生细菌的差异,以这两种栽培方式的菜心为材料,分离内生细菌.根据插入序列指纹图谱(IS-PCR)结果进行聚类,对代表性菌株的16S rRNA基因序列进行系统发育分析,并将菌株加入无土栽培营养液中,收获后测定菜心的的相对叶绿素含量(SPAD)和可溶性糖含量.结果显示,从两种菜心中共分离到31株内生细菌,IS-PCR结果将其聚为9个类群,16S rRNA基因鉴定其分别属于Bacillus sp.(芽孢杆菌属)、Rhizobium sp.(根瘤菌属)、Microbacterium sp.(微杆菌属)、Leifsonia sp.(雷夫松氏菌属)、Sphingomonassp.(鞘氨醇单胞菌属)、Alcaligenessp.(产碱菌属)和Agrobacteriumsp.(农杆菌属).土壤栽培菜心中可培养细菌数目多于无土栽培菜心,且土壤栽培菜心中内生细菌的碳代谢更多样化.生理生化实验显示,部分菌株能分泌生长素和铁载体.加入土壤栽培菜心中分离得到的菌株SCG14和SCG0802后,无土栽培菜心的相对叶绿素含量和可溶性糖含量比对照显著提高.本研究表明无土栽培和土壤栽培的菜心中的内生细菌在种类、数目、分布部位和碳源利用能力上有差异;此外,分离得到的菌株能提高无土栽培菜心的相对叶绿素以及可溶性糖含量,有开发为微生物制剂的潜力.(图4表6参35)  相似文献

Samples of soils underlying wastes were collected from four sites representing four demographic regions of a medium sized town in southwestern Nigeria. Standard methods and reference strains of isolated bacteria were employed for identification. Evaluation of the enzymatic and biochemical reactions showed that all isolated and identified microbes were non-fermenting heterotrophic (HTB). For example, Klebsiella pnemuniae may be involved in wound infections, particularly following bowel surgery. Similarly Pseudomonas aeruginosa can produce serious nosocomial infections if it gains access to the body through wounds or intravenous lines. From the 15 culure plates, 88 colonies with various characteristics were enumerated. They differed in aspect of viscosity and color. The bacterial species were identified by percent positive reactions while oxidative and sugar fermentation tests revealed various characteristics among the isolated strains. All of the isolates were negative for citrate utilization, gelatin liquefaction, nitrate reduction, methyl red and Voges Proskaur, motility and hydrogen sulphate production. The quantity of HTB present in an area serves as an index of the general sanitary conditions of that area. The presence of a large number of HTB, in an ecological area may be considered a liability as it can enhance the spread of diseases and on a larger scale may enable epidemics to arise. Therefore, there is need for control of waste sites by recovery and regular germicidal sanitation.  相似文献

从四川省豆科植物鸡眼草属、合欢属、豇豆属、葛属、金合欢、杭子梢分离获得了103株根瘤菌,采用AFLP、BOXAIR-PCR研究了这些菌株的遗传多样性.结果表明,分离自四川省豆科植物6个属的根瘤菌存在明显的遗传多样性,AFLP分析中,全部供试菌株的相似性为21%,全部供试菌株可分为26个AFLP遗传群;BOXAIR-PCR聚类分析结果表明,103个菌株分在70%相似性水平处,供试菌株被分为18个BOX遗传群.除部分菌株外,多数根瘤菌按宿主类型聚群,同一宿主而地理来源不同的根瘤菌分布于不同的遗传群,表明宿主和地理环境对根瘤菌具有深刻影响.图3表1参9  相似文献

The population of small copepod species (approximately <1 mm) were investigated during an annual cycle in Disko Bay, western Greenland. The small species considered were Acartia longiremis, Pseudocalanus spp., Oithona spp., Oncaea spp., Microsetella spp., and Microcalanus spp. Most of the small species were present in the surface waters year round and numerically dominated the community, and in biomass from late summer and throughout winter. Oithona spp. was numerically the main contributor, while Pseudocalanus spp. dominated in terms of biomass. In the uppermost 50 m, maximum abundance, biomass and secondary production were observed in late September after the phytoplankton production practically had terminated and the winter initiated. The free spawning Acartia longiremis showed a strong seasonal fluctuation in biomass and egg production, in contrast to the egg carrying species Pseudocalanus spp. and Oithona spp. These had a long spawning season and maintained a more stable biomass year round. Secondary production was estimated by three different ways: (1) based on the obtained specific egg production rates, (2) a temperature dependent equation, and (3) a multilinear regression taking temperature, body weight and chlorophyll into consideration. The contribution of the small species was insignificant when compared to the large Calanus species during the spring- and post-bloom. However, during late summer and winter, where Calanus had left the upper water strata for hibernation, the small species played a crucial role in the pelagic carbon cycling.  相似文献

Predicting and preventing outbreaks of infectious disease in endangered wildlife is problematic without an understanding of the biotic and abiotic factors that influence pathogen transmission and the genetic variation of microorganisms within and between these highly modified host communities. We used a common commensal bacterium, Campylobacter spp., in endangered Takahe (Porphyrio hochstetteri) populations to develop a model with which to study pathogen dynamics in isolated wildlife populations connected through ongoing translocations. Takahe are endemic to New Zealand, where their total population is approximately 230 individuals. Takahe were translocated from a single remnant wild population to multiple offshore and mainland reserves. Several fragmented subpopulations are maintained and connected through regular translocations. We tested 118 Takahe from 8 locations for fecal Campylobacter spp. via culture and DNA extraction and used PCR for species assignment. Factors relating to population connectivity and host life history were explored using multivariate analytical methods to determine associations between host variables and bacterial prevalence. The apparent prevalence of Campylobacter spp. in Takahe was 99%, one of the highest reported in avian populations. Variation in prevalence was evident among Campylobacter species identified. C. sp. nova 1 (90%) colonized the majority of Takahe tested. Prevalence of C. jejuni (38%) and C. coli (24%) was different between Takahe subpopulations, and this difference was associated with factors related to population management, captivity, rearing environment, and the presence of agricultural practices in the location in which birds were sampled. Modeling results of Campylobacter spp. in Takahe metapopulations suggest that anthropogenic management of endangered species within altered environments may have unforeseen effects on microbial exposure, carriage, and disease risk. Translocation of wildlife between locations could have unpredictable consequences including the spread of novel microbes between isolated populations.  相似文献

This research work was conducted in Uppanar estuary to ascertain the role of plasmids in the antibiotic resistance of bacteria. Water and sediment samples were collected for a period of three months. When tested against 20 antibiotics 22 MAR strains were isolated from the samples, which were found resistant to 5-13 antibiotics. They belong to 7 genera and 10 species. Gram-negative bacteria namely Neisseria mucosa, N. sicca, Branhamella catarrhalis, Klebsiella ozaenae, Citrobacterintermedius, Pseudomonas fluorescens and Enterobacter aerogenes were isolated. Gram-positive bacteria were of Bacillus subtilis, B. megaterium and Micrococcus luteus. When plasmid curing was done using acredine orange, the resistance against penicillin-G, ampicillin, tetracycline, amoxycillin, kanamycin, and chloramphenicol were totally lost in all strains, which confirmed the role of plasmid in these strains against antibiotics. Ten strains belong to different species were selected for the plasmid isolation and electrophoresis was done. Presence of plasmids in all strains was confirmed and the molecular weight was in the range of 2850 to 3170 bp. The study revealed that MAR strains are common in Uppanar estuary and they are plasmid mediated. This environment is seemed to be deteriorating at an alarming rate.  相似文献

Sensory and certain microbial analyses were applied to assess the quality of raw fish sold at a market in Siliguri cityof West Bengal, India. In regular surveys undertaken during June to August 2008, a particular fish species was randomly selected, its source was noted and a sensory analysis, the quality index method (QIM) was applied to assess its quality Raw fish samples were also collected and a small quantity (about 1 g) of scales oran upper layer of the skin surface (forscale-less fish samples), gill, liverand a portion of gut with gut-contents were aseptically removed for enumeration of the total aerobic heterotrophic bacteria, Aeromonas spp., Pseudomonas spp., Salmonella spp. and coliform counts. Oreochromis mossambicus and Tenulosa ilisha recorded significantly higher QIM scores, compared to other species (p<0.05). Riverine fish, Lepidocephalichthys guntea and Channa punctatus scored the lowest QIM scores (0) while scores for Puntius ticto and Mystus vittatus and pond cultured species like Cirrhinus mrigala, Catla catla, Labeo rohita, Labeo bata and Cyprinus carpio were very marginal (p<0.05). Aeromonas spp., Salmonella spp. and total coliforms were recorded from all the studied species while Pseudomonas spp. was isolated from only seven species. Among the tissues examined, the lowest counts of total heterotrophic bacteria, Aeromonas spp., Pseudomonas spp., Salmonella spp. and total coliforms were recorded from the skin in every fish species. Highest counts of pathogenic bacteria (except Pseudomonas spp.) were recorded in Tenulosa ilisha for all the tissues except liver. Since fish are properly cooked in Bengali households, the risk of disease from fish consumption is relatively less. However, some tribes residing in the region are known to consume undercooked fish and proper cooking methods should be followed in view of the present findings to avoid health risks. Risk Assessment of Riparian Plant Invasions into Protected Areas   总被引:6,自引:0,他引:6  
Abstract:  Protected areas are becoming increasingly isolated. River corridors represent crucial links to the surrounding landscape but are also major conduits for invasion of alien species. We developed a framework to assess the risk that alien plants in watersheds adjacent to a protected area will invade the protected area along rivers. The framework combines species- and landscape-level approaches and has five key components: (1) definition of the geographical area of interest, (2) delineation of the domain into ecologically meaningful zones, (3) identification of the appropriate landscape units, (4) categorization of alien species and mapping of their distribution and abundance, and (5) definition of management options. The framework guides the determination of species distribution and abundance through successive, easily followed steps, providing the means for the assessment of areas of concern. We applied the framework to Kruger National Park (KNP) in South Africa. We recorded 231 invasive alien plant species (of which 79 were major invaders) in the domain. The KNP is facing increasing pressure from alien species in the upper regions of the drainage areas of neighboring watersheds. On the basis of the climatic modeling, we showed that most major riparian invaders have the ability to spread across the KNP should they be transported down the rivers. With this information, KNP managers can identify areas for proactive intervention, monitoring, and resource allocation. Even for a very large protected area such as the KNP, sustainable management of biodiversity will depend heavily on the response of land managers upstream managing alien plants. We suggest that this framework is applicable to plants and other passively dispersed species that invade protected areas situated at the end of a drainage basin.  相似文献

In this study, a traditional assay was modified to evaluate the effect of Hg, Cd, and Zn on the bacterial community of a sequencing batch reactor and activated sludge plants and heavy metal-resistant bacterial species were determined. After the isolation of metal-resistant bacteria, their 16S rRNA gene fragments were sequenced. The BLAST program was used to compare the resulting 16S rRNA sequences with those in GenBank database to identify the isolated bacterial species. Hg was found to be the most toxic metal for both bacterial communities investigated. Sequence batch reactor bacteria were comparatively more resistant to Hg, Cd, and Zn than those from activated sludge. The resistant strains were close to the members of genus Pseudomonas, Kocuria, Stenotrophomonas, Enterococcus, and Staphylococcus. The modified dehydrogenase enzyme assay seems to be simple, robust, and competent for evaluation of the impact of metals on bacterial activity. Sequencing batch reactor systems should be preferred over activated sludge when wastewaters containing hazardous metals are to be treated.  相似文献

Vertical distribution, diet, and morphology of adults were examined in 27 species of euphausiids occurring in the upper 1000 m in the eastern Gulf of Mexico. Vertical distribution patterns were similar to those found in the central ocean gyres and oceanic equatorial waters of the Atlantic, Indian and Pacific Oceans. Most species migrated vertically from their daytime depths of 300 to 600 m to the upper 300 m at night. Exceptions were the non-migrating species of Stylocheiron, which remained in the epipelagic zone day and night, and Nematobrachion boopis, which remained in the mesopelagic zone. Based on gut-contents analysis, the Gulf euphausiids were largely zooplanktivorous, with cyclopoid and calanoid copepods being the most common items in stomachs. ostracods were especially common in the stomachs of Thysanopoda spp. and phytoplankton in the guts of Euphausia spp. Nearly every species' diet contained a considerable amount of olive-colored debris, which may have been marine snow generated in the epipelagic zone. Cluster analysis grouped the euphausiids into nine diet guilds. Euphausiids with a generalized morphology (i.e., spherical eyes, uniform thoracic appendages) tended to group together and demonstrated little variety in stomach contents among species. Euphausiids with a specialized morphology (i.e., bilobed eyes, elongate thoracic appendages) showed considerable variety in stomach contents among species, and several species had diets that were highly specific. Many of the species that had similar gut contents fed on prey of different sizes, as indicated by the width of the calanoid copepod mandibles found in stomachs. Principal-components analysis of seven morphological characters yielded species groups that were similar, but not identical, to those generated by cluster analysis of stomach contents data. We inferred from this that morphological characters partly determine diet, but that behavior is also important. Using the 20 most abundant species and 3 niche parameters, we attempted to identify the degree of separation among euphausiids based on the level of overlap in vertical distribution and diet composition, and on differences in mean prey size. Overlap of <60% in vertical distribution or diet composition was considered to indicate distinction of that parameter. Of 190 total species pairs, only 4 pairs did not demonstrate niche separation in at least one of these categories. We found that differences in these niche parameters were greatest among species with a specialized morphology and least among species that were morphologically generalized.  相似文献
