阐述了发光细菌的生理特性及其应用原理,重点评述了发光细菌法在工业废水、水域水质毒性及其他相关领域监测中的应用。与其他生物监测方法相比,具有快速、简便、灵敏等特点,测试结果与传统的鱼类毒性试验具有良好的相关性,实际应用方便可行。同时,结合现代电子技术和化学分析技术,对发光细菌法的发展进行了展望。  相似文献

比较了国产与进口HPLC-ICP-MS联用仪器在环境水样砷形态分析中的应用。国产和进口HPLC-ICP-MS联用仪器均能实现亚砷酸盐、二甲基砷、一甲基砷和砷酸盐的同时测定。国产仪器出峰时间较快,优于进口仪器。但是进口仪器上亚砷酸盐和二甲基砷的分离度优于国产仪器。国产仪器和进口仪器分析亚砷酸盐、二甲基砷、一甲基砷和砷酸盐,标准曲线的线性相关系数均大于0.995。国产仪器分析4种形态砷检出限范围为0.29~0.59μg/L,进口仪器分析4种形态砷检出限范围为0.24~0.36μg/L,没有数量级上的差异。国产仪器相对标准偏差范围为1.8%~9.0%,进口仪器相对标准偏差范围为0.8%~8.9%。精密度方面国产仪器和进口仪器水平相当。实际样品加标实验,国产仪器回收率范围为86.2%~112%,进口仪器回收率范围为96.4%~114%;使用进口仪器和国产仪器测定4种形态砷的有证标准物质,测定结果无显著性差异。国产HPLC-ICP-MS联用仪器可以满足砷形态检测分析的要求。对2款仪器的比较研究,为广大企事业单位及科研院所在选择HPLC-ICP-MS联用仪器时提供有价值的基础数据和参考建议。  相似文献

采用离子选择电极法测定水中氯化物,通过试验确定参比溶液、离子强度调节剂的配制,并用添加氯离子氧化剂和离子浓度调节剂的方法排除干扰离子对测定的影响,使得复合氯离子电极测得的能斯特曲线响应系数为93.2%。标准品的加标回收率为99.8%±0.6%,测定结果的 RSD为0.4%。用该方法与国标硝酸银滴定法同时测定实际水样,两方法的测定值绝对误差在允许范围内。  相似文献

中华人民共和国水利行业标准(SL 327.1-2005)和中华人民共和国城镇建设行业标准(CJ/T 51-2004)分别对原子荧光法测定水中砷作了一定的阐述,但两者对硫脲—抗坏血酸溶液的加入量有不同的规定,本文就这两种方法分别作了相应的实验,对两者的检出限、精密度以及加标回收率进行比较分析,得出两者在砷的测定方面没有显著差异。  相似文献

通过硝酸-过氧化氢微波消解样品,采用ICP-AES法测定了河水中锌、铅、镉、砷、镍、铁、铬、铜等重金属元素的含量,并分析了这些重金属元素在不同河段、不同水期含量的分布情况。测定结果的RSD<9%,加标回收率在86.0%~103.4%之间,检出限在0.832~2.42μg/L之间,该方法快速、稳定、效果较好。  相似文献

水样中只存在铵离子时对COD的测定几乎没有影响,而当铵离子与氯离子同时存在时,除了氯离子带来的干扰外,铵离子也会对COD的测定产生干扰。针对此现象,研究建立了加碱氮吹法,此法可有效去除COD测定中浓度小于1 000 mg/L铵离子的干扰,并确定了最佳除铵离子条件为过量加碱、600 m L/min的氮气流量和3 h的通气时间。通过7次平行测定含铵离子干扰物的水样,该方法的相对标准偏差为3.3%~6.5%,加标回收率为90.2%~97.0%,表明方法具有较好的精密度和准确度,能够较准确测定含有铵离子干扰水样的COD。  相似文献

选取不同类型的环境和污染源水样开展了发光细菌毒性测试,针对不同环境管理目标,探讨了急性毒性定量表征方式。发光细菌急性毒性测试在环境应急监测、水源早期生物预警中,采用绘制毒性预警基线图的方式比较可行。在污染源监督监测中,对于发光抑制率低于60%、无法求出EC50的样品,采用等效毒性参照物的质量浓度表征废水的毒性;对于发光抑制率高于60%,采用稀释因子表示样品的毒性更直观可靠。  相似文献

阐述了应用发光细菌检测有色水样毒性时,水样色度会引起附加发光抑制的原因,提出应用同心双层检测玻管,可以对有色水样色度引起的附加发光抑制加以修正.通过比较单层管和双层管测定标准曲线间的差异、设置阴性对照和阳性对照评价该方法的可靠性,以及用该方法检测有色水样活性黑KN-B溶液的急性毒性实验,结果表明,用同心双层检测玻管可以有效修正由水样色度引起的附加发光抑制.  相似文献

为了研究生物急性监测方法对监测典型化工污水处理厂废水的适用性,选择2家常州市典型的化工园区污水处理厂("常A"和"常B"),进行了发光细菌、藻类、大型溞和斑马鱼卵4种不同层次受试生物的急性毒性检测。研究表明,发光细菌急性毒性、藻类叶绿素荧光毒性在2个污水处理厂中均被检测到,发光细菌急性毒性通常进水大于出水,但在投放大量氧化消毒剂时,出水表现出剧毒。藻类叶绿素荧光毒性最高值出现在常B进水中。大型溞和斑马鱼卵急性毒性仅在常B进出水样中有检出,受纳河道下游水样虽未表现出急性毒性,但可观察到斑马鱼卵各类发育畸形。4种生物急性毒性检测方法中,发光细菌适用范围最广,藻类、大型溞和斑马鱼卵急性毒性方法可根据监测目的和工业园区特点相应选择。实验结论可为化工污水处理厂尾水排放过程中生物毒性监测的常态化提供方法选择,为管理部门制定排放标准提供依据。  相似文献

A biochemical oxygen demand (BOD) monitoring system, based on electrochemically-active bacteria in combination with a microbial fuel cell, has been developed for the purpose of on-site, on-line and real-time monitoring of practical wastewater. A microbial fuel cell that had been enriched with electrochemically-active bacteria was used as the basis of the measurement system. When synthetic wastewater was fed to the system, the current generation pattern and its Coulombic yield were found to be dependent on the BOD5 of the synthetic wastewater. A linear correlation between the Coulombic yields and the BOD5 of the synthetic wastewater were established. Real wastewater obtained from a sewage treatment plant also produced a highly linear correlation between the Coulombic yield and BOD5 in the system. To examine on-site, on-line and real-time monitoring capability, the BOD monitoring system was installed at a sewage treatment plant. Over 60 days, the measurement system was successfully operated with high accuracy and good stability with the measuring period for a sample being 45 min. This application showed that the application of the measurement system was a rapid and practical way for the determination of BOD5 in water industries.  相似文献

A multi-channel continuous water toxicity monitoring system was, after confirming the systems' performance, implemented to samples of water discharged from power plants to detect and classify their toxicity using several recombinant bioluminescent bacteria. Each channel of the system is composed of a series of two mini-bioreactors to enable a continuous operation, i.e., without system interruption due to highly toxic samples. A different recombinant bacterial strain was present in each channel: DPD2540 (fabA::lux CDABE), DPD2794 (recA::luxCDABE), and TV1061 (grpE::luxCDABE), which are induced by cell membrane-, DNA-, and protein-damaging agents, respectively. GC2 (lac::luxCDABE) is a constitutive strain, whose bioluminescence is reduced by an increase in cellular toxicity. Phenol and mitomycin C (MMC) were used for evaluating the system's performance to detect toxic chemicals. These samples were injected into the second mini-bioreactor according to a step or bell-curve manner. The field samples used in this study were obtained from the water discharged from two different power plants in Korea – from a nuclear power plant and a thermo-electronic power plant, and were injected into the second mini-bioreactor to initiate the toxicity test. Each channel showed specific bioluminescent (BL) response profiles due to the toxic compounds present in the water samples. Comparing the BL signals between the standard toxic chemical samples and discharged water samples, the equivalent toxicity of the field water could be estimated. Finally, it was proved that this novel continuous toxicity monitoring system can be used as an alternative tool for the quick monitoring and control of water quality, as well as aid in the setting up of a new monitoring strategy to protect the source of tap water and in the prevention of polluted water discharge.  相似文献

从2009年3月至2010年10月对京杭运河(苏州段)的4个监测断面进行了水质急性综合毒性和地表水主要理化项目的调查,结果表明:京杭运河(苏州段)水质急性综合毒性基本处在低毒水平,各断面基本呈现水质急性毒性水平枯水期>平水期>丰水期的规律,水质急性综合毒性测定结果基本与同步理化监测结果一致。  相似文献

Interests in the determination of different arsenic species in natural waters is caused by the fact that toxic effects of arsenic are connected with its chemical forms and oxidation states. In determinations of water samples inorganic arsenate (As(III), As(V)), methylated metabolities (MMAA, DMAA) and other organic forms such as AsB, AsC, arsenosugars or arsenic containing lipids have the most importance. This article provides information about occurrence of the dominant arsenic forms in various water environments. The main factors controlling arsenic speciation in water are described. The quantification of species is difficult because the concentrations of different forms in water samples are relatively lowcompared to the detection limits of the available analytical techniques. Several hyphenated methods used in arsenic speciation analysis are described. Specific advantages and disadvantages of methods can define their application for a particular sample analysis. Insufficient selectivity and sensitivity of arsenic speciation methods cause searching for a new or modifications already existing techniques. Some aspects of improvement and modifications of the methods are highlighted.  相似文献
