Usefulness of automated chromatography for rapid fetal blood analysis for second trimester prenatal diagnosis of β-thalassemia

Prenatal diagnosis of β-thalassemia is now ideally done in the first trimester of pregnancy by chorionic villus tissue DNA analysis. Nevertheless, fetal blood analysis in the second trimester is required either when the mutation in both parents cannot be characterised or when the couple comes late for investigations. We evaluated the usefulness of analysis of fetal blood on the Biorad Variant Hemoglobin Testing System using the β-thalassemia short programme in comparison with the conventional globin biosynthesis in 58 pregnancies. The β/α biosynthesis ratios in 13 homozygous fetuses ranged from 0 to 0.03 and the adult hemoglobin (HbA) levels by automated chromatography varied from 0% to 0.4%. The normal or heterozygous fetuses had β/α ratios of >0.04 and HbA levels ranging from 2.1% to 10.6%. In 17 fetuses we also correlated the β gene mutations with the predicted genotypes using automated high-performance liquid chromatography (HPLC). Effect of varying pH on protein composition and yield of amaranth seed (Amaranthus blitum)

The isolation procedure of the seed proteins of Amaranthus blitum have been analyzed at different pH conditions. Qualitative studies were carried out by using electrophoretic technique sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). Mainly four protein fractions i.e. albumin, globulin, prolamin and glutelin were obtained. Protein isolates were prepared by (a) extraction at different alkaline pH (9, 10, 11 and 12) and precipitation atpH 5 and (b) extraction at pH 9 and precipitation atdifferentpH (4, 5, 6 and 7). The composition of isolates, prepared by method (a), depended on extraction pH. The isolate extracted at pH 8 was mainly composed of albumin and globulin, whereas at pH 9, 10 and 11 showed the presence of prolamin and glutelin. Electrophoretic pattern of different isolates had five major bands with molecular weight of 30, 45, 72, 84 and 90 kDt respectively. The increase of the extraction pH led to the increase in protein yield. With method (b) isolates obtained were variable in composition. At pH 7 albumin and prolamine were present, whereas at pH 4, 5 and 6 all 4 protein fractions were precipitated. According to the quantitative estimation of the albumin, globulin, prolamin and glutelin in the seed flour the contents were 26.4, 25, 5.81 and 42.7%, respectively. The results suggest that composition of protein isolates could be controlled by different extraction and precipitation pH.     

Transforming time into money using water: A participatory study of economics and gender in rural India

When water supply improvements are coupled with opportunity to create income through micro–enterprises, time released from water collection is converted into income earned. This brings several benefits: reduced drudgery, higher household income, and, consequently, greater women's empowerment through changing gender relations within the household. This article documents the performance of one such scheme in Banaskantha District in the state of Gujarat in India, one of poorest districts in the state and the country. Here, due to the efforts of the Self–Employed Women's Association (SEWA), an Indian NGO, poor women are reaping the social and economic benefits of a government–run regional piped water supply scheme, a project funded by Dutch bilateral aid, focusing on women's development. If government policy–makers took the cue and formulated programmes and schemes that combined these two aspects, viz., actions that release time for rural women from daily chores (e.g., collecting water, fuel wood and fodder)and opportunities for sustainable micro–enterprises to convert time saved into income, this could become a reliable route out of rural poverty into gender–sensitive sustainable development.
