In the present study, biosynthesis of gold nanoparticles (AuNPs) by the cells (cells-AuNPs) and cell-free extracts (extracts-AuNPs) of a new fungus Mariannaea sp. HJ was reported. The as-synthesized particles were characterized by UV-vis spectroscopy, transmission electron microscopy (TEM), X-ray diffraction (XRD), and Fourier transform infrared spectroscopy (FTIR). The effects of different parameters on AuNP biosynthesis were investigated, and initial gold ion concentration of 2 mM, pH 7, was demonstrated to be suitable for both cells-AuNP and extracts-AuNP syntheses. The cells-AuNPs were of various shapes, including sphere, hexagon, and irregular shapes, with an average size of 37.4 nm, while the extracts-AuNPs were almost spherical and pseudo-spherical with an average size of 11.7 nm. XRD pattern suggested that the crystal structure of both AuNPs was face-centered cubic. FTIR spectra implied that some biomolecules from the fungal cell walls or cell-free extracts were involved in the formation of AuNPs. The as-synthesized AuNPs were demonstrated to have excellent catalytic activities for the reduction of 4-nitrophenol with the catalytic rate constants of 5.7 × 10?3/s for cells-AuNPs and 24.7 × 10?3/s for extracts-AuNPs. Desertification has emerged as a serious threat to the alpine meadows of Northwest Sichuan in recent decades. Artificial vegetation had certain effects on desertification recovery, while how the CO2 flux changed and its reasons are still unclear. During the growing season in 2016 (i.e., from July to September), we selected the desertified alpine meadows with different recovery degrees, including the early stage of restoration, the middle stage of restoration, the late stage of restoration, and control (the unrecovered desertification meadow) as four transects. CO2 flux was measured by the instrument LI-8100, and the microenvironment factors that affected CO2 flux changes were analyzed. The results showed that the carbon sequestration function of desertified alpine meadows gradually increased with the degree of recovery. Net ecosystem exchange (NEE) were -1.61, -3.55, and -4.38 μmol m-2 s-1 in the early, mid-term, and late transects, respectively, and the most dramatic changes occurred from the early stage to mid-term stage, increasing by 120.50%. Both ecosystem respiration (ER) and soil respiration (SR) were enhanced significantly with restoration (P < 0.05). In mid or late July, NEE, ER, and SR reached their maximum values, and thereafter, the indicators varied to near zero (P < 0.05). During the whole growing season, the daily dynamic in CO2 flux for the control alpine meadow was mild and retained the trend of continuous release all day, but that in the desertified alpine meadow was a single peak pattern. Moreover, with restoration process, the peak of CO2 flux increased and reached a peak in the late stage of the recovery process. The regression analysis showed that there was a significant positive correlation between CO2 flux and vegetation coverage, aboveground biomass, and soil moisture (0-5 cm) (P < 0.01), and a weak correlation with 0-5-cm soil temperature (P < 0.01). This indicates that topsoil moisture (5 cm) is a more significant factor for CO2 flux than topsoil temperature during the growing season in the restoration of desertified alpine meadows in Northwest Sichuan. In general, the vegetation recovery significantly improved the carbon-sequestration ability of the desertified alpine meadows during the growing season in Northwest Sichuan, and at the middle stage of restoration, the carbon-sequestration ability improved significantly due to vegetation restoration and increase in topsoil (0-5 cm) moisture. © 2018 Science Press. BACKGROUND: The Yangtze Delta is one of the most developed regions in China and includes Shanghai, eight cities in Jiangsu province and eight cities in Zhejiang province. Meat consumption in this region has increased with economic growth, and most of the consumed meat is produced locally. The water quality of surface waters has deteriorated in recent years. An example was the huge blue-green algae bloom in Tai Lake in late May 2007, which affected millions of people's daily drinking water. However, animal husbandry is considered to be one of the main pollution sources. METHODS: Pollutants (NH3-N, total phosphorus (TP), and total nitrogen (TN)) excreted by livestock and poultry, and the resultant COD (chemical oxygen demand) and BOD (biochemical oxygen demand), were estimated using two different methods based on different data sets. RESULTS: The number of livestock and poultry has remained stable in the Yangtze Delta over the four years from 1999 to 2002, with the average number of pigs, cattle, sheep and poultry being 21.1 M, 0.4 M, 7.7 M and 597.6 M, respectively. Pollutants in livestock and poultry excreta estimated by Method I were: 0.12 Mt NH3-N, 0.11 Mt TP and 0.29 Mt TN, resulting in COD and BOD of 1.34 Mt and 1.30 Mt, respectively, while the estimations based on Method II were: 0.18 Mt NH3-N, 0.15 Mt TP and 0.40 Mt TN, resulting in COD and BOD of 1.95 Mt and 1.80 Mt, respectively. DISCUSSION: Pollutants excreted annually by livestock and poultry in the Yangtze Delta are estimated to be: 0.17 Mt NH3-N, 0.16 Mt TP and 0.42 Mt TN, giving rise to a COD of 1.86 Mt and a BOD of 1.72 Mt. Approximately 25% of this pollution was estimated to enter water bodies, which means that the annual pollutant load is 43,700 t NH3-N, 39,400 tTP, 104,600t TN with a COD of 465,000 tand a BOD of 430,100 t. Pollutants from animal husbandry were similar in magnitude to those from industrial wastewater. Pigs produced the most pollution, followed by poultry, cattle and sheep. The pollution load from animal husbandry in the Yangtze Delta is about twice the average level of the whole of China. CONCLUSIONS: Domestic wastewater was the main pollution source in the Yangtze Delta, followed by pollution from raising livestock and poultry and from industrial wastewater. The pollution load in Shanghai and Jiaxing were the greatest, followed by 7 cities of Jiangsu province (except Suzhou) and other cities of Zhejiang province and Suzhou. Pigs and poultry produced about 90% of the total pollutants from animal husbandry. RECOMMENDATIONS AND PERSPECTIVES: The local governments, especially in Shanghai and Jiaxing, should focus their attention on the pollution produced by livestock and poulrry. Controlling pollution from pigs and poultry will have the greatest impact in this region. The chemical characteristics of fine particulate matter (PM2.5) emitted from commercial cooking were explored in this study. Three typical commercial restaurants in Shanghai, i.e., a Shanghai-style one (SHS), a Sichuan-style one (SCS) and an Italian-style one (ITS), were selected to conduct PM2.5 sampling. Particulate organic matter (POM) was found to be the predominant contributor to cooking-related PM2.5 mass in all the tested restaurants, with a proportion of 69.1% to 77.1%. Specifically, 80 trace organic compounds were identified and quantified by gas chromatography/mass spectrometry (GC/MS), which accounted for 3.8%–6.5% of the total PM2.5 mass. Among the quantified organic compounds, unsaturated fatty acids had the highest concentration, followed by saturated fatty acids. Comparatively, the impacts of other kinds of organic compounds were much smaller. Oleic acid was the most abundant single species in both SCS and ITS. However, in the case of SHS, linoleic acid was the richest one. ITS produced a much larger mass fraction of most organic species in POM than the two Chinese cooking styles except for monosaccharide anhydrides and sterols. The results of this study could be utilized to explore the contribution of cooking emissions to PM2.5 pollution and to develop the emission inventory of PM2.5 from cooking, which could then help the policymakers design efficient treatment measures and control strategies on cooking emissions in the future. 相似文献

毒性评估是人体健康风险评价的重要部分,目前主要存在2种方法,一种基于PAHs相对于苯并[a]芘(BaP)的毒性当量因子,采用Ba P致癌斜率因子参数(方法 1),另一种直接采用各PAHs致癌斜率因子和非致癌参考剂量等参数(方法 2)。然而2种毒性评估方法得到的风险及修复量是否存在差异以及引起差异的原因等问题鲜有讨论。针对苏南某焦化厂PAHs污染土壤,采用分层土壤健康风险评价模型,对比了2种毒性评估方法确定的PAHs风险、修复目标污染物及土方量的差异,并对引起差异的关键因素进行探讨。结果表明:对于0.0~1.0 m表层土壤,方法 1和方法 2确定的致癌风险最大值分别1.48E-05和1.32E-05,均超过可接受致癌风险,修复土方量分别为27 846 m~3和28 667 m3,修复目标污染物均为Ba P和二苊烃(Acy)。对于1.0~3.0 m深层土壤,方法 1确定的致癌风险最大值为3.36E-08,低于可接受致癌风险,不需要修复;而方法 2确定的致癌风险最大值为3.73E-04,非致癌危害指数最大值为6.96E+01,分别超过可接受致癌风险和非致癌危害商,需要修复,修复目标污染物为萘(NaP),修复土方量为35 944 m~3。最终,方法 2确定的总修复土方量(64 611 m~3)为方法 1确定土方量(27 846 m~3)的2.45倍,而这种差异主要是由于方法 1低估了深层土壤中高挥发性PAH单体尤其是NaP的风险所致。因此,从保守角度建议采用方法 2进行PAHs风险评价。 相似文献

为了明确内质网应激(endoplasmic reticulum stress,ERS)介导的凋亡在2,2’,4,4’-四溴联苯醚(2,2’,4,4’-tetrabromodiphenylether,PBDE-47)致大鼠神经毒性中的作用,在脑发育的突增期(出生第10天),分别用0 mg·kg~(-1)、1 mg·kg~(-1)、5 mg·kg~(-1)和10mg·kg~(-1)PBDE-47进行单次灌胃染毒,并从出生第8天开始每天给予150 mg·kg~(-1)ERS抑制剂4-苯基丁酸(phenylbutyric acid,PBA),持续3周。仔鼠出生第8周末,从对照组、10 mg·kg~(-1)PBDE-47处理组、150 mg·kg~(-1)PBA处理组和150 mg·kg~(-1)PBA+10mg·kg~(-1)PBDE处理组中随机选取8只大鼠进行水迷宫实验。然后将所有动物断头处死,分离大鼠海马组织,观察其海马组织形态学改变、测定ERS标志分子(GRP78、IRE1和CHOP)和凋亡相关蛋白Cyt c的表达水平。结果显示,10 mg·kg~(-1)PBDE-47处理可导致雌性大鼠海马CA4区细胞排列紊乱、锥形神经细胞数量减少甚至消失,尼氏小体数量减少,大鼠逃避潜伏期延长(P0.05)。5和10 mg·kg~(-1)PBDE-47处理可显著上调ERS相关蛋白IRE1和CHOP的表达水平(P0.05),并明显增强海马组织凋亡相关蛋白Cyt c的表达。PBA干预可明显降低PBDE-47诱导的IRE1和CHOP以及Cyt c蛋白的表达水平,提示PBDE-47可通过ERS介导凋亡,导致大鼠海马组织损伤,从而影响大鼠学习记忆功能。 相似文献
