The rapid development and increase of antibiotic resistance are global phenomena resulting from the extensive use of antibiotics in human clinics and animal feeding operations. Antibiotics can promote the occurrence of antibiotic resistance genes (ARGs), which can be transferred horizontally to humans and animals through water and the food chain. In this study, the presence and abundance of ARGs in livestock waste was monitored by quantitative PCR. A diverse set of bacteria and tetracycline resistance genes encoding ribosomal protection proteins (RPPs) from three livestock farms and a river were analyzed through denaturing gradient gel electrophoresis (DGGE). The abundance of sul(I) was 103 to 105 orders of magnitude higher than that of sul(II). Among 11 tet-ARGs, the most abundant was tet(O). The results regarding bacterial diversity indicated that the presence of antibiotics might have an evident impact on bacterial diversity at every site, particularly at the investigated swine producer. The effect of livestock waste on the bacterial diversity of soil was stronger than that of water. Furthermore, a sequencing analysis showed that tet(M) exhibited two genotypes, while the other RPPs-encoding genes exhibited at least three genotypes. The sigma (SIG) coordinate system in ocean circulation simulation models results inevitably in horizontal pressure gradient error. This problem also emerges in models of deep lakes or reservoirs with the same characteristics of underwater terrain mutation. SIG coordinates reflect vertical relative stratification but cannot be used to calculate horizontal pressure gradient force in places with drastic topographic changes; this results in vertical water temperature and circulation errors. In deep lakes or reservoirs, differences in water density caused by the temperature difference between upper and lower water bodies is the primary cause of thermal stratification phenomena. Lake Mead was used as a case study on steep topography based on Environmental Fluid Dynamics Code (EFDC) model in this study. SIG coordinates result in close agreement between the calibrated temperature time series at the top and middle water layers, but disparity in the bottom water layer. The error emerges in the horizontal pressure gradient error due to the SIG coordinate transformation. Neither increasing the vertical resolution nor adjusting the horizontal viscosity coefficient resolve this error. We test the sigma-zed (SGZ) coordinate which combines Z coordinate and SIG coordinate as a replacement for the SIG coordinate to find that they effectively reduce the model’s runtime and simulation efficiency. The vertical temperature distribution in SGZ coordinate mode is more accurate than the distribution in SIG coordinate mode. The Navier-Stokes horizontal gradient and advection diffusion equation results under SIG coordinates are very sensitive to the pressure gradient. The replacement also enhances resolution near the thermocline, facilitates reclosing of the water bottom and the equal sigma surface, lends significant advantages in terms of vertical temperature in the simulation for local deep water with steep terrain, and shortens runtime for 0.14 h. SGZ mixed coordinates are recommended in the simulation of deep lakes or reservoirs wherein the underwater topography is large (with abundant continuous deep trenches or reefs).

Biodegradation of polycyclic aromatic hydrocarbons (PAHs) is very difficult in saline-alkaline soil due to the inhibition of microbial growth under saline-alkaline stress. The microorganisms that can most effectively degrade PAHs were screened by introducing microorganisms immobilized on farm byproducts and assessing the validity of the immobilizing technique for PAHs degradation in pyrene-contaminated saline-alkaline soil. Among the microorganisms examined, it was found that Mycobacterium sp. B2 is the best, and can degrade 82.2% and 83.2% of pyrene for free and immobilized cells after 30 days of incubation. The immobilization technique could increase the degradation of pyrene significantly, especially for fungi. The degradation of pyrene by the immobilized microorganisms Mucor sp. F2, fungal consortium MF and co-cultures of MB+MF was increased by 161.7% (P < 0.05), 60.1% (P < 0.05) and 59.6% (P < 0.05) after 30 days, respectively, when compared with free F2, MF and MB+MF. Scanning electron micrographs of the immobilized microstructure proved the positive effects of the immobilized microbial technique on pyrene remediation in saline-alkaline soil, as the interspace of the carrier material structure was relatively large, providing enough space for cell growth. Co-cultures of different bacterial and fungal species showed different abilities to degrade PAHs. The present study suggests that Mycobacterium sp. B2 can be employed for in situ bioremediation of PAHs in saline-alkaline soil, and immobilization of fungi on farm byproducts and nutrients as carriers will enhance fungus PAH-degradation ability in saline-alkaline soil. 相似文献
